Purpose:1. Association between ATP1A2 Exon23 SNP/G772A, Intron8 SNP/A57G and Intron20 SNP/T1842G Gene Polymorphisms and elite aerobic performance.2. Association between ATP1A2 Exon23 SNP/G772A, Intron8 SNP/A57G and Intron20 SNP/T1842G Gene Polymorphisms and the effects of endurance training.Subjects and Methods:Subjects:①EEA(elite endurance athletes): They comprise 123 unrelated Chinese elite endurance athletes. They were specialists in long-distance running including 5000m, 10000m and marathon.②SCON(sedentary control): They comprise 206 unrelated Chinese sedentary undergraduates. They studied in Beijing Sport University.③NS(new soldiers): They consist of 102 unrelated Chinese man soldiers from a police army. The SCON and the NS have no trained experiences, as well as their parents and grandparents. All subjects are Han ethnicity and plain-men in North China (including Liaoning,Heilongjiang,Jilin,Hebei,Tianjin and Shandong province).Methods: There are tow parts in the study including the case-control study and the association study. PCR-RFLP was used to examine the genotype and allele frequencies of Exon23 SNP/G772A, Intron8 SNP/A57G and Intron20 SNP/T1842G Gene Polymorphism of the three subjects. Find the association between aerobic performance of the elite endurance athletes and these gene polymorphisms. The NS undertook a distance running program consisted of 5000 m running with the intensity of 95-105% ventilatory threshold (VT), 3 times per week for 18 weeks. VO2max,running economy (RE),body composition and Left ventricular structure and function were measured before and after the protocol, and were used to explore the association between ATP1A2 (ATPapse, Na+/K+ transporting,alpha-2polypeptide) gene polymorphisms and the effects of endurance training. X2 test was used to compare the frequencies between different group subjects, and to determine whether the genotype distribution differed from that expected under the Hardy-Weinberg equilibrium. ANOVA (LSD) tested differences in variables before and after the training between the three genotypes. P<0.05 were considered statistically significant. All statistical analyses were performed with SPSS software for Windows 10.0 package. The results are presented as means±SD.Conclusions:1. ATP1A2 gene BglⅡexon21-22 polymorphism was at the 772bp in exon23 and was SNP G/A, and there were significant differences between in Chinese Han Ethnicity and those in Europe and America.2. GG genotype of G772A can become one of the gene polymorphism signals in selecting elite men 5km and 10km running athletes.3. G772A Gene Polymorphism contribute to individual response to endurance training for RE and Left ventricular function. 4. Our findings demonstrated that there were significant differences in allele and genotype frequencies between Chinese Han Ethnicity and European and American, but no significant difference was detected between Chinese population and Japanese population.5. AG genotype of A57G might become one of the gene polymorphism signals in selecting elite 5km and 10km running athletes.6. There were significant differences in allele and genotype frequencies between Chinese Han Ethnicity and European and Africa-Americans.7. TT genotype of T1842G might become one of the gene polymorphism signals in selecting elite women marathon athletes.
