Objective:To explore the role of NF-kB in exhaustive exercise-induced myocardial micro-injury, we studyed the distribution and variation discipline of NF-kB in different time by establishment of rat exercise-induced myocardial micro-injury model by single exhaustive swimming and repeated exhaustive swimming.Methods:In this study, we selected immunofluorescence staining and RT-PCR semi-quantitative techniques to explore the distribution and variation of NF-kB in the level of protein and mRNA in 24 hours after exercise, by establishment of rat exercise-induced myocardial micro-injury model.Results:Control group NF-κB mainly distributed in cytoplasm, cell membrane, tunica intima, occasionally in cell nucleus, but after exhausted exercise, NF-κB mainly distributed in cell nucleus, cell membrane and tunica intima.1,The content of NF-κB in left ventricle 6hours after single bout of exhaustive exercise was significantly higher than the control group, 12-hour group,24-hour group (p<0.05); the content of NF-κB p65 protein at 0 hours after exercise were significantly higher than control group (p<0.05), very significantly higher than the 12-hour group,24 hours (p<0.01) in Interventricular septum, and after exercise the 6-hour group was very significantly higher than 12-hour group (p<0.01); the content of NF-κB p65 protein at 0 hours after exercise was significantly higher than the quiet control group, and 24-hour group in right ventricle (p<0.05), and the 6-hour group was significantly higher than the control group (p<0.05).There was no significant difference between groups of 2-week repeated exhausted exercise groups in left ventricle and right ventricle. The content of NF-κB p65 protein in 12-hour group was significantly higher than 24-hour group in Interventricular septum (p<0.05).12-hour group after 2-week repeated exhausted exercise groups was very significantly higher than the 12-hour group after single bout of exhaustive exercise (p<0.01).2,The total content of NF-κB p65 protein in 0-hour group after single exhausted exercise was significantly higher than control group (p<0.05), 6-hour group very significantly higher than control group(p<0.01), 12-hour group significantly lower than 0-hour and 6-hour group.There was no significant difference between groups of 2-week repeated exhausted exercise groups of the total content of NF-κB p65.The total content of NF-κB p65 protein in 12-hour group after single exhausted exercise was significantly higher than 2-week repeated exhausted exereise groups (p<0.05) There was no significant difference between other groups.3,The content of NF-κB p65 mRNA of 0-hour group after single exhausted exercise was significantly higher than 24-hour group (p<0.05). The 6-hour group was significantly higher than the control group,24-hour group (p<0.05), and 12-hour group was significantly higher than 24-hour group (p<0.05).The content of NF-κB p65 mRNA of 6-hour group,12-hour group after repeated exhausted exercise was significantly higher than control group (p<0.05).The 6-hour group was significantly higher than the 0-hour group,24-hour group after 2-week repeated exhausted exercise.Conclusion: Both single bout of exhaustive exercise and repeated exhausted exereise can make the content of NF-κB protein and mRNA increase, and NF-κB could adjust the expression of many kinds of cell factors, and the damage caused by cell factors which adjusted by NF-κB may be the one of the important mechanism of cardiac muscle's damage led by exhaustive exercise.
