Objective: Bone tissue extracellular matrix is important for maintaining the structure and function of normal bone tissue. Collagen I is an important protein that constituted the bone tissue extracellular matrix. MMP-1 is a collagenase that mainly degraded collagen I. In the domain of sports medicine, there are not many researches about the effect and of exercise to collagen I and MMP-1. In this experiment, we investigate the effect and mechanism of collagen I adaptation to different exercise through the adaptation to 8 weeks treadmill exercise by biochemistry and molecular biology techniques.Methods:8-week-age SD big female rats (30) were grouped randomly into 3 groups in this experiment, the control group (n=10), the medium intensity exercise group (n=10) and the great intensity exercise group (n=10), the medium intensity exercise group and the great intensity group were trained separately 8 weeks. Test methods and targets:â‘ The expression of protein collagen I and MMP-1 with immunity histochemistry methodâ‘¡The expression of collagen I and MMP-1 in mRNA were analyzed by RT-PCR technique.Results: (1) expression influence of COL-I and MMP-1mRNA in rats bone tissue.â‘ Medium intensity exercise group expression COL-ImRNA was higher than the control group's in rat bone tissue(P<0.05),but the MMP-1mRNA expression level had tendency of enhance and non-statistics difference.â‘¡Great intensity exercise group in rat bone tissue expression of COL-ImRNA obviously increased, and was higher than the control group's and the medium intensity exercise group's . Conclusion: (1)collagen I increased for different intensity treadmill exercise. (2)The medium intensity exercise was helpful to bone collagen I content, but the great intensity exercise did not favor the bone collagen I content. (3)MMP-1 with different intensity treadmill exercise could play the influential role in degrading collagen I, degeneration ability of bone tissue extracellular matrix could affect the bone tissue adaptation to exercise.
