Effect Of Electroacupuncture At Neiguan Point On ATP - Sensitive Potassium Channel Associated Protein In Rats With Myocardial Ischemia

Purpose:The model of myocardial ischemia (MI) rats had been made by injecting isoproterenol (ISO) in this experiment, while observed the protein expressions of Kir6.1, Kir6.2, SUR2A, SUR2B on the ATP-sensitive potassium channel and PKA, PKC, PKG as the auxiliary proteins of ATP-sensitive potassium channel. The aim of this experiment was to explore the possible mechanism and the meridian specificity of improving MI and hypoxia by acupuncture.Material and method:According to a table of random numbers, ten rats from 72 adult, healthy, male SD rats were drawn at random as the control group and were each given multiple point subcutaneous injections of normal saline solution in the medial limb roots. Then the electrocardiograph (ECG) was connected. The remaining 62 rats were injected with ISO (85 mg/kg) in the same manner. Both groups of rats were injected twice after 24 h and then another ECG was recorded. 40 rats were included in the study, which were divided into model group, Neiguan group, Lieque group and neither meridians nor points group. Each group contained 10 rats. "Hua Tuo brand" acupuncture needles (0.20 mm X 25 mm) were inserted into each corresponding point 2 mm under the skin. A 68054) electro-acupuncture (EA) device was connected and rats were subjected to a density wave (2-20 Hz) until stable. Current intensity was judged appropriate when forelimbs appeared consistent with a slight quiver of EA frequency. Rats were stimulated for 20 min once a day, seven times consecutively. Non-acupuncture treatments were done on both the control and model groups. The protein expressions of Kir6.1, Kir6.2, SUR2A, SUR2B and PKA, PKC, PKG were analyzed by western blot.Results:1. Compared with the control group, the T-wave voltage of rats’ECG significantly decreased in MI (P<0.05). Compared with the model group, the T-wave voltage of Neiguan and Leique groups increased, respectively (P<0.05) and the effects of Neiguan group were higher than Lieque group. T-wave voltage of rats’ECG did not change significantly in the neither meridians nor points group (P>0.05). 2. Compared with the control group, the protein expressions of Kir6.1, Kir6.2, SUR2A and SUR2B significantly increased in MI (P<0.05). Compared with the model group, the protein expressions of Neiguan and Leique groups decreased, respectively (P<0.05) and the effects of Neiguan were higher than Lieque. The protein expressions of neither meridians nor points group did not change significantly (P>0.05). Compared with the Lieque group, the protein expressions in Neiguan group significantly decreased (P<0.05).3. Compared with the control group, the expressions of PKA, PKC and PKG significantly increased in the model group (P<0.05). Compared with the model group, the expressions of Neiguan and Leique groups decreased, respectively (P <0.05). The expressions of neither meridians nor points group did not change significantly (P>0.05). Compared with Neiguan group, the expressions in Lieque group significantly increase (P<0.05).Conclusion:1. T-wave voltage decreased significantly in MI rats. It proved that EA at Neiguan and Lieque could decrease the increase, but the effect of EA at Neiguan was better than Lieque.2. The protein expressions of Kir6.1, Kir6.2, SUR2A and SUR2B were low in health rats, but they increased significantly in MI rats. EA at Neiguan and Lieque could decrease the increase effectively, and the effect of EA at Neiguan was better than Lieque.3. The expressions of PKA, PKC and PKG as ATP-sensitive potassium channels’ auxiliary proteins increased significantly in MI rats. EA at Neiguan and Lieque could decrease the increase effectively, and the effect of EA at Neiguan was better than Lieque.4. Neiguan of pericardium meridian and Lieque of Lung Meridian were located in the inside of forearm near the wrist position, which can improve the protein expressions of ATP-sensitive potassium channel and its auxiliary proteins. All the results proved the meridian specificity of Neiguan.
