Purpose:To observe the effect of electro-acupuncture at Neiguan point on the subunit protein expression of ATP-sensitive potassium channels(SUR2A, SUR2 B, Kir6.1, Kir6.2) and expression of protein kinase(PKA, PKC-β) of ASIC3-/-myocardial ischemia mice.Material and method: 40 ASIC3-/- healthy male 8-week-aged mice(C57BL/6)were supplied by Beijing Weitong-Lihua laboratory animal Corp. Ltd. These mice were randomly divided into blank group, model group, Neiguangroup, Lieque group, Zusanli group and Non-acupoints group. Formal experiment was launched after adaptive feeding lasting one week. All mice were given ISO(20mg/Kg) in method of intraperitoneal injection to establish myocardial ischemia mice model, except for the blank group. Record the electro cardio gram(ECG) of all mice before and after modeling. Except for the blank control group, other groups were given electro acupuncture treatment according to their group. The treatment were given daily and lasted for 7 days. Observe the change of ECG and the expression of protein kinase and ATP-sensitive potassium channels by the method of western blot.Result:1. After the electro acupuncture treatment, compared with the model group, the T-wave amplitude of Neiguan, Lieque and Zusanli group were significantly increased. The group without meridians and acupoints has no statistic meaning and the T wave amplitude of Neiguan group has no disparity with it in Lieque group and Zusanli group.2. After the electro acupuncture treatment, compared with the model group, the protein expression of ATP-sensitive potassium channels(SUR2A, SUR2 B, Kir6.1, Kir6.2) was significantly decreased.3. After the electro acupuncture treatment, compared with the model group, the protein expression of protein kinase(PKA, PKC-β) was significantly decreased.Conclusion:1. Electro-acupuncture on Neiguan, Lieque and Zusanli acupoints can increase the T wave amplitude of myocardial ischemia mice obviously.2. Electro-acupuncture on Neiguan acupoint can decrease the expression of ATP sensitive potassium channels protein SUR2 A, SUR2 B, Kir6.1 and Kir6.2 in cardiac muscular tissue of myocardial ischemia mice.3. Electro-acupuncture on Neiguan acupoint can decrease expression of protein kinase(PKA,PKC-β) in cardiac muscular tissue of myocardial ischemia mice.4. On mice with myocardial ischemia, Neiguan-point is better than Lieque-point and Zusanli-point. Neriguan’s regulation of Kir6.2, SUR2 A and PKA is the reflection of specific acupoints along meridians.
