Purpose:ASIC3 gene was chosen as the experimental target to knock out(ASIC3-/-)mice, myocardial ischemia model was built by isoproterenol medicine to inquire into the influence of relative protein Kv1.4、Kv4.2、Kv4.3 and auxiliary protein’s expression in outward ion channel on different acupuncture points to inquire into the mechanism of acupuncture to myocardial ischemia. Meridians special characteristic of improvement in myocardial ischemia was tested and proved from it.Method:Health male ASIC3-/- mice was fed in good condition, six of which was chosen as the control group. The rest was injected subcutaneous isoproterenol(ISO) into abdomen(20 mg/kg)twice every 24 hours,meanwhile, the blank group was injected same amount of normal Saline. With the measurement of cardiogram, we compare the alteration before and after the treatment model. 30 ASIC3-/-mice, model built already, were divided into model group, PC6 group, LU7 group, ST36 group and non-meridian and non-acupoints group with 6 in each group randomly. “Hua Tuo” brand acupuncture needles were pricked into PC6 group, LU7 group, ST36 group and non-meridian and non-acupoints group about 2mm while they were connected to electric acupuncture apparatus, adjusted to dilatational wave with 2m A for 20 min retaining needles, 7days continuously. After the acupuncture, we adopt Western blot to inspect the expression of Kv1.4、Kv4.2、Kv4.3 and auxiliary protein KCh IP2.Result:1. Compared with the control group, T wave in model group’s electrocardiogram had apparently pathological alteration which owned statistical meaning(P<0.01). T wave of mice electrocardiogram in PC6 group,LU7 group and ST36 group all had improvement after 7days acupuncture in which alteration range had statistical meaning(P<0.05), but there was still disparity in voltage valves of rats’ wave, while the T wave of mice electrocardiogram in non meridians and acupoints group had little alteration which had no statistical meaning(P>0.05).2.Compared with the blank group, the expression of outward ion channel protein Kv1.4、Kv4.2、Kv4.3 and auxiliary protein KCh IP2 decreased which had statistical meaning(P<0.01). Compared with the model group, the index of protein expression in PC6 group had higher voltage valves which was better than the LU7 group and ST36 group 7 days after electroacupunture and the disparity had statistical meanings(P<0.05). While the index of protein expression in the PC6 group had higher voltage valves,obviously better than the LU7 group and ST36 group(P<0.05)and the index of protein expression in the non meridians and acupoints group had little alteration(P>0.05).Conclusion:1.Electro-acupuncture at PC6,LU7, ST36 could obviously improve the ECG T-wave abnormalities when myocardial ischemia happened in mice, and of the three points, PC6 was the best.2. The expression of the channel-associated protein of Kv1.4、Kv4.2、Kv4.3、KCh IP2 will decreased in myocardial ischemia mice, However, by electro-acupuncture PC6、LU7、ST36could significantly increase the expression of these proteins,especially which performed better in PC6.3. PC6 point belongs to the Pericardium Meiridian which close to LU7 that belongs to the Lung Meridian.the two points above have a regulatory function to the cardiac transient outward potassium channel; ST36 has the same effect through its comprehensive capabilities. However, data was shown that PC6 specificity in the treatment of myocardial ischemia.
