The Effect Of Acupoints On The Expression Of Transient Outward Potassium Channel-related Genes In Cardiomyocytes Of ACCN3 -/- Knockout Mice

Purpose:In this experiment,ACCN3-/-gene knockout mice as the research object,with isoprenaline hydrochloride(ISO)mouse model of myocardial ischemia induced by electroacupuncture in mice,Neiguan,Zusanli,Lieque,Nonpoints Kv1.4 on myocardial cells of experimental mice,transient outward potassium ion channel genes Kv4.2 and Kv4.3.The auxiliary subunits of KCh IP2 m RNA expression,from gene expression levels to explore mechanism of meridian acupoints acupuncture on myocardial ischemia,observation on the effect of pain transmission by transfer.Material and method:The ACCN3-/-gene breeding SPF adaptive knockout mice 52,were randomly selected 8 rats as control group.Intraperitoneal injection of normal saline in control group mice,the remaining mice were injected with isoprenaline hydrochloride(ISO),to induce myocardial ischemia in mice model by injection for two consecutive days.4 mice died in the modeling process,the remaining 40 mice were randomly divided into model group,Neiguan group and Zusanli group,Lieque group,non acupoint group 5 group,8 rats in each group.The experimental mice were fixed awake,in addition to the blank group and the model group were not treated with electroacupuncture stimulation the.Four groups of mice were given electroacupuncture intervention correspondingly,1 times a day,continuous stimulation for a week.Use of "Hua Tuo" brand into the corresponding acupoint acupuncture depth is 2mm,connecting electroacupuncture sustained 20 min stimulation,the stimulation intensity is 2m A frequency of 4 Hz/20 Hz Shu dense wave.ECG after stimulation respectively recorded 5 mice.After the experiment,in the experiment center in the mice were killed and promptly drawn,the mice were killed after the broken ridge,in the ice quickly isolated heart tissue,ice saline rinse,cut atrium and large blood vessels,cut along the left and right ventricular septum.Every one of the EP tube with the corresponding number,will be placed in the heart tissue of EP corresponding to the tube,add Trizol reagent 1ml per just within each EP tube by liquid nitrogen freezing,immediately stored in ultra-low temperature refrigerator(freezer temperature at-80?),waiting for the measurement experiment.Using real time quantitative polymerase chain reaction(Real-Time PCR)technology,to observe the myocardial cell of mice on Kv1.4,Kv4.2,Kv4.3,KCh IP2 expression of m RNA,the use of medical statistics software measurement index.Results:1.Compared with the blank group,the expression of Kv1.4,Kv4.2,Kv4.3,KCh IP2 m RNA in the other four groups of myocardial cells was significantly reduced,and the change was statistically significant(P < 0.05,P < 0.01).2.Compared with the model group,Neiguan group,Lieque group,Zusanli group,myocardial cell Kv1.4,Kv4.2,Kv4.3,KCh IP2 m RNA expression increased significantly,statistically significant change(P < 0.05,P < 0.01),the change of non acupoint group was not statistically significant(P > 0.05).3.Compared with Neiguan,Lieque group,Zusanli group,non acupoint group myocardial cells of Kv1.4,Kv4.2,Kv4.3,KCh IP2 m RNA expression decreased,significant change(P < 0.05,P < 0.01).4.Compared with the Lieque group,non acupoint group myocardial cells of Kv1.4,Kv4.2,Kv4.3,KCh IP2 m RNA expression decreased,significant change(P < 0.05,P < 0.01),Zusanli group change was not statistically significant(P > 0.05).5.Compared with the Zusanli group,non acupoint group myocardial cell Kv1.4,Kv4.2,Kv4.3,KCh IP2 m RNA expression decreased,difference of variation was statistically significant(P < 0.05,P < 0.01).Conclusion:1.Zusanli,Neiguan,Lieque by acupuncture can effectively resist myocardial ischemia.2.Anti myocardial ischemia effect of Neiguan Zusanli and superior Lieque point.3.Neiguan point on myocardial ischemia with meridian specificity.The sensation of meridians is not affected by pain.
