Objection With the model of fat-1 mice, we investigated the effct of the enriched endogenous ω-3 polyunsaturated fatty acids(PUFA) on the effect of body weight and probiotic bacteria, and then explored the mechanism of the phenomena,.Methods Heterozygous transgenic fat-1 mice, which came from Professor Jingxuan Kang of Harvard Medical College and Jianbo Wan of University of Macau, were mated with C57BL/6 wild-type mice to obtain wild-type(WT) and fat-1 littermates. All of the 6 weeks old healthy male mice were randomly divided into two groups: transgenic mice group and normal mice group.Body weight, length and fat weight were recorded per week, during the trail period. At the end of the experiment, we killed the mice and weighed the mice visceral around fat. Blood samples were taken to determine the content of the blood glucose(BG), triglyceride(TG), total cholesterol(TC). Fresh fecal matter and hypothalamus were got from the mice. NPY, CART, Ag RP, POMC, Nestifatin-1 and Ghrelin m RNA in hypothalamus were measured by q PCR.The expressions of bifidobacterium and lactobacillus in fresh facal matter were measured by q PCR. The expressions of LC3 and P62 were measured by western blot.Results The body weights, fat weights, BG, TG and TC in fat-1 mice were lower than that of the WT mice. Compared with wild-type mice, the m RNA levels of CART, POMC and Ghrelin were higher, while the m RNA levels of NPY, Ag RP and Nesfatin-1 were lower in fat-1 mice. Probiotic bacteria, including bifidobacterium and lactobacillus, were increased in fat-1 mice. The expression of LC3II/I was higher in fat-1 mice than that of WT mice; while Sequestosome-1( P62)was lower.Conclusion ω-3 polyunsaturated fatty acid dehydrogenase in fat-1 mice, which can confer ω-6PUFAs to ω-3 PUFAs, increased the endogenous ω-3 PUFAs and reduced the ω-6 /ω-3 ratio. Therefore,the body weights in fat-1 mice were reduced, the probiotic bacteria were improved.
