Fresh engineering flies maggots were used as experimental material and pH 8.0 phosphate buffer was used to extract flies maggots protein.Protein, total sugar, fat, total nitrogen and amino nitrogen content was determined respectively, the molecular weights distribution of proteins was estimated by SDS-PAGE. Experimental results showed that the extraction of flies maggots contained protein 9.88 mg/mL, fat content was 0.24 mg/mL, total sugar content was 0.25 mg/mL, total nitrogen content was 0.5 mg/mL, amino nitrogen content was 1.23 mg/mL. There were nine protein bands with the molecular weights of 91.8 kD,58.9 kD,42.9 kD,41.9 kD,33.0 kD,23.9 kD,18.2 kD,16.9 kD,16.4 kD identified using SDS-PAGE. The protein component of 91.8 kD was in the majority of the total proteins analyzed by gel imaging software.Fermentation was exploited with flies maggots extraction by Aspergillus oryzae Huniang 3.042. The protease activity of fermented liquid for 72 h was the highest,the main protein component was consumed during the transformation. After fermenting, the fermented liquid protein revealed eight new proteins with the molecular weights of 65.9 kD,49.8 kD,49.2 kD,44.7 kD,29.8 kD,25.8 kD,25.6 kD,22.4 kD respectively. Tricine-SDS-PAGE was run and some small peptides of 5.9 kD,5.0 kD,4.3 kD were detected.The content of 96 h fermented peptides is 7.89 mg/mL by biuret methed.Antioxidant activity assay in vitro showed the clearance rate of 25 mg/mL of 96 h fermented liquid on hydroxyl free radicals and superoxide anions achieved 88.79 % and 95.45 %, 22.72 % and 28.04 % higher than that of control.Anti-aging ability fermented liquid for 96 h was compared with flies maggots extraction in vivo using Drosophila melanogaster as experimental animals. Results showed the high dosage of fermented liquid for 96 h prolonged the average life-span of the Drosophila melanogaster.The avreage life-span of female Drosophila melanogaster was increased 29.15 %, it was increased 35.82 % for male.The high dosage of flies maggots extraction prolonged the average lifes-pan of the Drosophila melanogaster. The avreage life-span of female Drosophila melanogaster was increased 26.66 %,it was increased 31.73 % for male, indicating fermented liquid for 96 h had significant antioxidative and anti-aging effects.
