Cordyceps sinensis is a fungal parasite on the larvae of Lepidoptera, belongs to the family Clavicipitaceae of the order Hypocreales. Cordyceps sinensis is one of the most valuable medicinal fungi in traditional Chinese medicine. It possesses a variety of biologically active components, such as cordycepin, polysaccharides, ergosterol, etc. Cordyceps polysaccharides, one of its richest functional components, have many pharmacological features, such as immune activity modulation, hypoglycemic effect, and antitumor activity, etc. However, natural production of the fungus is very limited, so the research for substitutes of the natural Cordyceps sinensis by artificial culture develops quickly. In this paper, the Cordyceps sinensis mycelium was cultured by using agricultural products, through solid state fermentation. Optimized by single factor experiments and orthogonal method test, the selection of the fermentation substrates and optimization of culture condition was achieved.The gross and the rare of the Cordyceps sinensis mycelium were compared in the liquid seed fermentation. Through the selection of the fermentation substrates and optimization of culture condition, finalizing the liquid medium and culture condition is glucose 1.0%, yeast extract powder 1.5%, MgSO4·7H2O 0.15%, KH2PO4 0.15%, pH at 6.0, broth content at 40%(v/v), cultured at 22℃for six days, and the rate of the mycelium would reach 32%.In the solid state fermentation experiment, the results focus on the mycelium output. By comparing several kinds of agriculture products and optimizing culture condition, finalizing A 60% and B 40% as raw material, with X 3%. Through single factor experiments and orthogonal test, the optimum conditions were as follows:45 g of the filling capacity,25% of inoculation,125% of water content,22℃of temperature,12 days of fermentation time. Mycelium production is 7.17% in optimal conditions.Intracellular polysaccharide was extracted by hot water extraction. The best ethanol concentration was determined by using Sephadex G-100 gel chromatography through grading trend deposit. Then, we optimized the extraction processes of the Cordyceps sinensis polysaccharide by single factor experiments and orthogonal test. According to the results, the extraction processes were determined as follows:85℃of extraction temperature,1/100 of solid-liquid ratio, and 3.5 h of extraction time. The extraction rate of the solid-culture Cordyceps sinensis polysaccharide is about 2.4% in the optimal conditions. The infrared spectra of the solid-culture polysaccharides show that it contains absorption peaks of saccharide, mainly are the O-H stretching vibration and C-H stretching vibration, as well as weak C=C stretching vibration.
