The present article describes several extraction processes were used respectively toextracte the active constituents from the wide and growed S.divaricata,differentmethods and different polar solvent. The high-performance liquid chromatographic wasundertaken in order to determine the compounds content. Results indicated that thechromones content from wide S.divaricata is higher than the one from growedS.divaricata. The reflux extraction method was the optimum extraction. And80%ethanol extraction solvent was the optimum extraction.The present article developed high-performance counter-current chromatography(HPCCC) and rapid preparation chromatography for the separation and purification ofthe chromones from S.divaricata,and found to be a fast and efficient tool for separationof chromones from S.divaricata. On the one hand,four chromones were isolated andpurified from S.divaricata by high-performance counter-current chromatography(HPCCC) using a system consisting of ethyl acetate/n-butanol/ethanol/water(1:1:0.1:2,v/v/v/v). Each at over90%purity as determined by high performance liquidchromatography (HPLC). On the other hand,six chromones were isolated and purifiedfrom S.divaricata by HPCCC and rapid preparation chromatography. At last,thestructures of compounds were identified by their retention time, the liquidchromatography-electrospray ionization-mass spectrometry (LC-ESI-MS) in thepositive ion mode, and confirmed by NMR. The characteristic LC-ESI-MSfragmentation patterns of the four compounds were discussed,and found to be a veryspecific and useful tool for the structural identification of chromones from S.divaricata.The ethanol extracts from S.divaricata were subjected different methods for the evaluation of their antioxidant activity,respectively. Folin-Ciocaileu method wasdeveloped for evaluation of the total phenolic content (TPCs) of different extracts. TheTPCs of80%ethanol extract at90min showed the highest TPCs. The free radical2,2-diphenyl-1-picrylhydrazyl (DPPH˙) assay was then used to measure the antioxidantactivity of different extracts. The80%ethanol extract at120min exhibited strongantioxidant activity.
