Eugenol is the main component of clove essential oil. At present, the reportsabout eugenol are concentrated on the antimicrobial activity and anesthetic action onfish. Few repots study the antioxidant activity of eugenol in vivo and viable cell. Theobjective of this study is to investigate the antioxidant activity in vitro and thepreventive effect of eugenol on acute mice liver damage and cell oxidation damage,explore the antioxidant activity and mechanism of eugenol. The result of this studywill act as theory reference for the application of eugenol in food additive andfunctional food.The results of the antioxidant activity of eugenol in vitro indicated that reductionability of eugenol was significantly (P<0.05) high compared to BHT at theconcentration range of100~250μg/mL. The half·OH radical scavenging dose ofeugenol was152.52μg/mL which is lower than BHT. The DPPH scavenging rate ofeugenol with concentration of250μg/mL reached92.5%which is significantly(P<0.05) highe compared to BHT. The half O2-· radical scavening dose of eugenol was215.99μg/mL. The inhibition activity of eugenol on yolk lipoprotein system oxidationhas not significant difference compared to Vc at the concentration range from50μg/mL to200μg/mL. The inhibition activity of eugenol with concentration of250μg/mL on yolk lipoprotein system oxidation was significantly (P<0.05) high comparedto Vc. The antioxidant activity of eugenol in vitro increased significantly (P<0.05)with the increasing in concentration level of50~250μg/mL.To explore the preventive effect of eugenol on CCl4-induced liver injury in Balb/cmice, establish liver injury models acute by intraperitoneal injection of CCl4after thelast administration, all the mice were sacrificed to assay antioxidant activities in theblood and liver. Meanwhile, hepatic histopathological changes were also examined.Compared with model group, high dose (15mg/(kg·d)) and middle dose (10mg/(kg·d))eugenol can significantly (P<0.01) reduce blood AST, ALT activity, inhibition ofhepatic MDA content, and enhance liver SOD, GSH-Px, CAT, GSH-Px and CAT vitality activity, eugenol can alleviate mice liver inflammation. Eugenol can protectagainst CCl4-induced acute liver injury in mice by eliminate oxygen free radicals,protect the liver cell membrane integrity.HL7702Oxidative damage was induced by H2O2, the infuence of eugenol onH2O2-induced HL7702cell was assessed by measuring the superoxide dismutase(SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) activities andmalondialdehyde (MDA) content. It was found that the addition of10mmol/L ofeugenol has the best effect on the proliferation of HL7702growth. Eugenol withconcentration level of0.1~10mg/L can significantly (P<0.05) increase SOD, CAT,GSH-Px activity of HL7702, reduce the content of oxidation products of MDA.
