Su Ma is a traditional medicinal plant,rich in many nutrients.Guizhou hemp germplasm resources are rich and widely cultivated.The study found that the oil content of Guizhou Suma seed is about 30%,the linolenic acid content of up to about 60%,the unsaturated fatty acid up to about 90%,so Suma seed is to supplement the body of essential ?-3 polyunsaturated fatty acid plant and regulating fatty acid nutrition balance ?-3 polyunsaturated fatty acid(omega-3PUFAs)nutrient source.?-Linolenic acid is an essential fatty acid in human body.?-Linolenic acid deficiency is an important cause of dementia,cancer,cardiovascular and cerebrovascular diseases,hypertension,hyperlipidemia and diabetes.Recent studies showed that alpha linolenic acid and long-chain metabolites may play an important role in many physiological activities,such as intelligence,protect eyesight,reducing blood fat,lowering blood pressure,inhibit platelet aggregation,anti thrombosis,anti-aging,anti allergy and inhibit the occurrence and metastasis of cancer.In this paper,two different methods were used to isolate and purify alpha linolenic acid from Guizhou Suma which is a special plant resources.The effects of four factors,including distillation temperature,preheating temperature,rotating speed and feed rate on purification efficiency of molecular distillation were studied by single factor experiment.The optimum technological conditions of the reaction surface are: the distillation temperature is 100 ?,the preheating temperature is 60 ?,the rotational speed is 235 r/min,and the feed rate is 0.9 mL/min.At this time,the purity of the ALA in the collecting bottle is 83.01%.In order to obtain high quality and high purity of alpha linolenic acid,a multi-stage distillation experiment was carried out to control the distillation temperature.The purity of ALA was increased to 86.04% after the distillation of the 4 stage.For separation and purification of Guizhou Suma oil in alpha linolenic acid by ultrasonic assisted beta HP-?-CD inclusion method,considering the ultrasonic power,time and mixed fatty acids(ethanol +FFA)ratio and HP-?-CD solution(the ratio of material to water)three factors of alpha linolenic acid and purity.On the basis of single factor optimization design response surface.The results showed that under the condition of ultrasonic power of 250 W,the optimum extraction process was 1:7.5,and the extraction time was 21 min.Under this condition,the purity of alpha linolenic acid was about 87.11%.Animal experiments,continuous extraction in mice in a month of alcohol and molecular distillation process of different purity of alpha linolenic acid,analysis of serum apolipoprotein and related indexes of antioxidant enzymes in the liver and anti-inflammatory immune factors.The results show that the alpha linolenic acid intervention significantly reduced serum alanine aminotransferase(ALT),alkaline phosphatase(AKP)and total bilirubin level,can effectively promote the synthesis and secretion of apolipoprotein,inhibition of tumor necrosis factor(TNF-),interleukin(IL-6)and NO release.About 86% of alpha linolenic acid has the best effect on liver and liver,and the mechanism of action is to prevent the metabolism of triglyceride and cholesterol,the way of anti inflammation and antioxidation.The mice were fed with different purity of alpha linolenic acid for 10 days,and then injected with concanavalin A for a single time.Liver injury was assessed by measurement of serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)and tumor necrosis factor(TNF-),IFNand IL-6 levels.The levels of MDA,SOD,GSH,MPO in liver tissue were measured to determine the protective effect of alpha linolenic acid on the liver injury induced by concanavalin A in mice,and to explore the mechanism of action.The research shows that alpha linolenic acid has a protective effect on ConA induced liver injury in mice,the mechanism is to reduce proinflammatory cytokines,restore redox balance,thereby reducing the signal of liver cells and tissue damage.In this paper,a new method for the purification of alpha linolenic acid in crude oil from two kinds of sesame oil was studied.Different purity alpha purified by molecular distillation method of linolenic acid and explore its protective effect on alcoholic liver injury,concanavalin A induced liver injury in mice,found that alpha linolenic acid(especially high purity)has great potential in protecting liver and preventing immune liver injury.
