In Chinese culture, peony represents happiness, wealth and fortune. It is been widely planted in China with its high value of ornamental and medicine use. The root of peony is called "Danpi" in traditional Chinese medicine that is highly effective in detoxification, lowering blood pressure and removing blood stasis. It also can be used for the treatment of surgical diseases and skin diseases, anti-bacterial and anti-inflammatory clinical applications.The economic value of peony mainly stems from its ornamental and medicine use, but its usage is limited due to the backward technology of the deep processing after the flowering stage. In order to solve this, recently many researchers have focused on the peony seed. The extracts of peony seed contain37kinds of compounds, and17kinds of them are fatty acid, which most are unsaturated fatty acids. The highest content of it is linoleic acid, linolenic acid and oleic acid. Linoleic acid and linoleic acid have effects with the treatment of cardiovascular diseases, promoting fat decomposition and accelerates the rate of metabolism, anti-cancer, promoting the growth and development, promoting bone growth, and enhance the body’s immune function. It has high economic value.This paper established the evaluation method of the extraction process efficiency. It is evaluated by oil extraction rate and quantitative results of unsaturated fatty acids.In this paper, we optimize gas chromatographic separation conditions of fatty acid methyl ester, and inspect it with the methodology. The experiment results prove that gas chromatogram of palmitic acid, oleic acid, linoleic acid and linolenic acid are up to baseline separation in this chromatographic separation conditions. In this given circumstance, the result listed below. The linear relationship of oleic acid, linoleic acid and linolenic acid is accurate (R2=0.9992,0.9959and0.9986). The reproducibility of oleic acid, linoleic acid and linolenic acid are good (RSD=0.78%,0.77%and0.61%). The average recovery of oleic acid, linoleic acid and linolenic acid are99.6%,99.6%and99.8%(RSD=0.57%,0.34%and0.5%). This method is sensitive, accurate and reliable and suitable for the assaying of oleic acid, linoleic acid and linolenic acid in peony seed oil.Secondly using the single factor experimental design and response surface to optimize the extraction process of peony seed oil. Because of the main unsaturated fatty acids are oleic acid, linoleic acid and linolenic acid in peony seed three kinds of, and unsaturated fatty acids are of great value in health and nutrition, so by yield efficiency and oleic acid, linoleic acid, linolenic acid content of three kinds of unsaturated fatty acid as the evaluation standard of peony seed oil of microwave aided extraction process is optimized, So, the optimal extracting conditions are microwave power800W, ratio of material to liquid9mL/g, extracting times8mins. The contents of linolenic acid, linoleic acid, oleic acid are42.6%,29.1%,23.3%respectively, and the contents of all unsaturated fatty acids are94.2%, the oil extraction rate is27.7%.This paper also applies the gas chromatographic technique to test the four common edible fatty acids of soya-bean oil, tea oil, olive oil and colza oil with with peony seed, finding that the peony seed oil contains the largest amounts of unsaturated fatty acids, especially in linoleic acid and linolenic acid, which indicates its greater economic and health value.
