Peony has a unique scent,distinctive color at the same time it has many biological activities can not only for food but also medicinal.And the economic value of peony is also very high.The scientific research of peony is primarily to test and evaluation the effective components and volatile oil in the flower.At present,the studies of peony seed oil are rarely reported both at home and abroad.The peony seed oil as the fewer research parts so far,has the very high nutritional value.The oil yield efficiency of peony seed is between 25%and 38%,GC-MS detection result show that peony seed oil contains a variety of fatty acid compositions.The unsaturated fatty acids accounted for more than 80%,and saturated fatty acids accounted for only about 14%,while the unsaturated fatty acids base on the linoleic content of 22.19%,27.14%oleic acid and about 27.14%linolenic acid.Therefore peony seed oil is a kind of ideal health daily cooking oil.On March 22,in 2011,our country's health ministry has approved the peony seed oil as a new resource food,for the high value applications of peony seed oil opened a new-chapter.The peony seed oil has above advantages,how can we make better applications of peony seed oil?This article afforded a new method and new idea for the peony seed oil extraction,storage,purification and further applications,it can ensure that the main nutritional compositions of peony seed oil are well preserved and maximum utilization.The main conclusions are as follows:First of all the single factor method is adopted to define the main influence factors of microwave extraction of peony seed oil and scope of main factors were determined:used the mixed solvent of ethanol and n-hexane as extraction solvent,selected 20 mesh particle size for microwave extraction experiments.Set the main factors influencing scope as:extraction temperature 70-100?,extraction time 2-10min,ethanol ratio in the mixed solvent 10-20%,solid-to-liquid ratio 0.14-0.5g/mL.In order to more detailed exploration of the influence law of various parameters on the yield and energy consumption in the microwave extraction process of peony seed oil,we design response surface optimization experiment with the Design Expert 8.0 software.Through the experiment to get the best extraction conditions as follow:solid-to-liquid ratio 0.33g/mL,extraction time 2.86min,extraction temperature81.41 ? ethanol ratio 20.00%,theory of optimal extraction yield 34.06%.According to the above,the optimal conditions were repeated three times peony seed oil extraction average yield is 33.98%on average,good repeatability and lowest energy dissipation.The peony seed oil which is separate extracted by soxhlet extraction,solvent extraction,supercritical fluid extraction and microwave extraction were all analyzed by using GC-MS,the results show that the peony seed oil which was extracted by four different extraction methods unsaturated fatty acid content is basically the same,but microwave extraction of minimum energy consumption,the shortest time,the operation is relatively simple,peroxide value and malondialdehyde of microwave extract peony seed oil were relatively low,the acid value was almost the same with other extraction peony seed oil.In 0.02%amount of various antioxidant addition,the condition of the influence of oxidation stability by various antioxidants for the peony seed oil storage is:TBHQ>rosemary extract>bamboo extract>VE.As synthetic antioxidants antioxidant effect of TBHQ is most obvious,and antioxidant effect of rosemary extract is optimal in nature antioxidants.The condition of the influence of malondialdehyde by various antioxidants for the peony seed oil storage is:TBHQ>rosemary extract>bamboo extract>VE.Malondialdehyde value of blank group increased from 0.41 mg/kg to 5.25 mg/kg and TBHQ and rosemary extract group malondialdehyde increased to 1.39 mg/kg and 2.14 mg/kg,far lower than the blank group.Peony seed oil storage experiments confirmed that the temperature,illumination,air will promote the peony seed oil oxidation,of which the influence of different temperature for the peony seed oil oxidation is most remarkable,the influence of light is after temperature and the influence of air is relatively small.For prolonged storage and ensure its nutritional value of peony seed oil,it should be transported and deposited at low temperature and sealed away from light environment.We get the optimized preparation condition of the peony seed oil ?-cyclodextrin inclusion by single factor optimization method.The optimized preparation condition of ?-CD inclusion peony seed oil to preparation of peony seed oil curing powder are as follows:inclusion temperature 50 ?,the ratio of ?-linolenic and ?-CD mole number is 1:2,inclusion time is 2.5 h,the amount of ethanol is 4.5 times the dosage of peony seed oil(w/w).The optimal inclusion rate is 69.93%,oil content rate is 26.74%.SEM results:after ?-CD inclusion of peony seed oil the surface is smooth,closely and close connection between molecules is not prone to deformation and fracture,etc.,which can determine the peony seed oil can be tightly wrapped inside the ?-CD formed "host-guest" molecular structure.1H NMR and FT-IR,the peony seed wrapped compound still retain the original cavity structure of ?-CD 's main mapping features,and compared with FTIR of the peony seed oil,inclusion and peony seed oil spectrum structure difference is bigger,its external distribution of the ?-CD.XRD results show that the crystal type show a basic change of inclusion less crystalline structure and the neat arrangement is more closely.DSC results show that melting point of inclusion relative reduced to the ?-CD the inclusion reaction,which is in accordance with the testing results of XRD and SEM.TGA results show that,the peony seed wrapped compound structure stability of weightlessness condition similar to ?-CD,the similarity can reach 70-80%.The compound solid powder of Peony seed oil maximum concentration is 1.33 times more than that of peony seed oil group.The AUC value of compound solid powder of Peony seed oil is 2200.641 mg/L*h,the AUC value of the peony seed oil is 2018.33 mg/L*h,so it means that bioavailability of compound solid powder is higher than that of peony seed oil.Using cyclodextrin package solvent method and extraction method purify alpha linolenic acid of peony seed oil.And we use the Box-Behnken model by the Design Expert 8.0 software optimize purge process.The finally optimized conditions is:the inclusion temperature 59.5?,the inclusion time 3.75 h,the mole ratio of FAEE&?-CD 0.31,FAEE:Ethanol dosage for FAEE is 3.2 times.The best preparation conditions for the purification of samples in FTIR and GC detection.GC results confirm the purified alpha linolenic acid purity can reach 76.04%.SEM showed ?-CD and purification of alpha linolenic acid structure more compact surface,smooth and stable.FTIR show peony seed oil congealing clathrate mainly reflects the characteristics of the structure of ?-CD,and combined with GC detection results show that peony seed oil entered into the ?-CD inside the cavity.
