Marasmius androsaceus is a Chinese traditional medicinal fungus, which has pharmacological,edibleness and health care functions. So the study of Marasmius androsaceus liquid submergedfermentation and its pharmacological activities has important significance and value.This experiment optimized Marasmius androsaceus fermentation conditions through the singlefactor experiment and orthogonal experiment, And on this basis, determinated the antioxidant activities. Onintracellular and extracellular polysaccharide of Marasmius androsaceus, and applied mice experiments toanalyse the analgesia and sedation pharmacological functions of Marasmius androsaceus fermentationproducts. The main research contents and results of experiments are as followings:The medium and fermentation conditions of Marasmius androsaceus were optimized to improvethe biomass of Marasmius androsaceus. Through screening the type of C source and N source andoptimizing the concentration of C source and N source, a orthogonal experiment was used to optimize themedium component of Marasmius androsaceus. The optimal medium is established as: corn flour:3%,wheat bran:3%, MgSO4:0.1%, KH2PO4:0.3%, VB1:10mg, water:1000mL; By screening fermentationtime, temperature, speed, initial pH of the medium, inoculum concentration, and volume of liquidinoculum, the optimal fermentation conditions are established as: fermentation time:6d, temperature:25℃, rotation speed:150r/min, initial pH:5, inoculum size:9%, liquid volume:90mL,The mycelium and fermentation broth of Marasmius androsaceus were made by fermenting.using alcohol precipitation method extracted extracellular polysaccharide from fermentation broth. Using water extraction and alcohol precipitation method extracted the intracellular polysaccharide frommycelium. With ascorbic acid as control, through the experiment in vitro the antioxidant activities of theintracellular and extracellular polysaccharide of Marasmius androsaceus has been studied on scavenginghydroxyl, suoeroxide (·O-2) anion and1,1-Dipheny-2-picrylhydrazyl(DPPH) radicals. The results showethat extracellular polysaccharide and intracellular polysaccharides have significant scavenging ability onhydroxyl and DPPH, and the ability to clear the hydroxyl radical is intracellular polysaccharide>extracellular polysaccharides. However, the ability to scavenge DPPH is extracellular polysaccharides>intracellular polysaccharide.There is no notable scavenging effect on superoxide anion. The scavengingability of polysaccharide of Marasmius androsaceus is significant in three different free radical scavengingsystem in vitro, it shows that Marasmius androsaceus has strong antioxidant activities.In hot-plate test, determined pain threshold of mouse in the time of1hour after druged, theresults showe that the pain threshold of mouse after administration are longer than the pain threshold ofmouse before administration and physiological saline group, the difference is statistically significant.Control group and medium concentration group have the better effects, the results indicate that Marasmiusandrosaceus have analgesia function. In tail suspension test, the silent period of mouse are shorten afternitroglycerin molding, which has difference to compared with physiological saline group. The silent periodof mouse are length after administration, the difference is statistically significant. The diazepam group andmedium concentration of Marasmius androsaceus have clearly effects, the results indicate that Marasmiusandrosaceus have sedation function.In toxicological experiment, Marasmius androsaceus fermentation which dosage amounts to200times than clinical medicine were administered to mice, the mice doesn’t appear the phenomenon as abnormal activity and dead in three days. Compared with the experiment group and control group, there isno significantly different about the increase of mice weight, the results indicate that Marasmiusandrosaceus almost have no toxic and side effect.
