Marasmius androsaceus is a traditional Chinese medicinal fungi, It is mainlyused for treating traumatic injury, fractures pain, sciatica, migraine, leprosyneuralgia and rheumatic arthritis in our country folk. In the industrial scaleproduction, The raw material is mainly obtained from the solid culture method ofMarasmius androsaceus mycelium fermentation. Research shows that the mainchemical constituents of Marasmius androsaceus are mannitol, cholesteryl acetate,amino acids, hydroxycinnamic acid, ergosterol, and plenty of carbohydrates andprotein components etc..In our study,we used two kinds of chemical mutagenesis method forimproving on Marasmius original strain: two ethyl sulfate and sodium nitritemutation breeding. from the multi-level screening, we obtained a strain ofgenetically stable, excellent strains, named T08. Compared with the original strain,its mycelium dry weight increased by36.40%. At the same time, the geneticstability results of high yield performance the strain of T08shows, it could bestably inherited. In addition, this chapter studied on the confirmation of theeffective components of the obtained mutant strain T08from Marasmius, itproved that the chemical constituents of fermented mycelia of products containsamino acid, the polysaccharide, polypeptide, phenols, lactone, grease, terpenoids,alkaloids and saponins etc..Considering the dry weight of high yield strain T08fermentation mycelium,intracellular polysaccharides, mannitol and adenosine production,we applicatedthe satisfaction function to the fermentation of Marasmius medium andfermentation pot culture conditions in the process of exploration. Sequentiallythrough the single factor experiment, Box-Behnken experimental design, multipletwo regression analysis and artificial neural network combined genetic algorithmmode,we optimized the medium components and culture conditions offermentation tank of fermentation of Marasmius high-yielding strain T08. Theresults showed: the best culture medium formula of Marasmius high-yield strain(g/L): sucrose23.1, peptone10, yeast extract10.4, magnesium sulfate seven water1, Sanshui0.86potassium dihydrogen phosphate, and vitamin B10.1; the best5Lfermentation conditions: initial pH6.14, speed289r/min, fermentation time6.28 days, fermentation temperature of26DEG C,5%inoculation quantity, age4days,ventilation volume200L/h, liquid volume3.5L/5L, compared to the initialculture conditions, the dry weight of mycelia fungus, adenosine, polysaccharideand mannitol production are also increased respectively by35.02%,23.34%,187.05%and74.45%.The analgesic activity study of the high yield of Marasmius T08:throughinduced writhing test by acetic acid, Faure Marin induction experiment and hotplate experiment, Marasmius analgesia groups are preliminary screened; throughself activity and balance the rotarod test, we studied if the ethanol extract of themycelia of body has neurotoxicity; Through several antagonist screening,monoamine component level determination and Western blot experiments wereused to determine the possible mechanisms of analgesic effect of ethanol extract.Ultimately, determined the body of ethanol extract from Marasmius androsaceusexerting the analgesic ability was by inhibiting the synthesis of dopamine, theeffect was similar to the dopamine synthesis inhibitor.The study on antidepressant activity in high-yield Marasmius T08: by thespontaneous activity of mice experiment, balance the rotarod and early safety tests,it determined the Marasmius metabolic polysaccharide has a certain degree oftoxicity; Through the behavioral despair models in the forced swimming test andthe tail suspension test it preliminarily determined the anti-depression Ability tometabolic polysaccharide; Through5-HTP and RES induced mechanism model,hypothalamus monoamine level changes and Western blot experiments todetermine the metabolic polysaccharide exerts antidepressant effects similar topossible; through t he rat model of chronic unpredictable stress model,we studythe changes of body weight, partial addicted to sugar water, forced swimming test,tail suspension test, real monoamine level changes and Western blot experimentsto further explore similar antidepressant effect of metabolic polysaccharide mayexist. The results showed, Marasmius T08fermentation metabolic polysaccharidecan play a certain effect of anti depression, The effect is related to its regulation ofcatecholamine neurotransmitter systems and consisted with the onset ofdepression monoamine hypothesis.
