Lemon fruit contains many kinds of bioactive components,and its bioactivities areabundant. With the rapid development of economic and improvement of people’s quality oflife, more and more researchers pay attention to the lemon. In the studies on bioactiveconstituents, anticancer, anti-senlity, antioxidant, reducing blood sugar etc.With the figuresrelated to the use of lemon, rapid propagation technique for improvin-g fruit quality becomethe inevitable course in the planting industry. The research and development of health careproducts of lemon is on the upsurge. This thesis on the lemon propagation techniques werestudied preliminary, and the total dietary fiber, lemon polyphenols, flavonoids, limonoids, thecontent of mainly metal elements were classified, and bioactive peptides were studied.In order to detect the dietary fiber in lemon, the enzymatic gravimetric method, thecontent of protein and ash was determined by Kjeldahl and ashing method, then the dietaryfiber in lemon homogenate were determined quantitatively. Result was21.9mg/g, theprecision and repeatability performance is good, the relative standard deviation of7.01%.In order to research the total polyphenol in lemon, Folin-Ciocalteu was used to determinethe content of total polyphenol. When the concen-tration was0.1mg/mL-1.0mg/mL the linearof polyphenol standard is good, the standard curve of R2=0.999. The result was1.224mg/mL,the standard recovery and repeatability of the extract which was diluted250times,50times,25times were good, RSD:8.43%,2.27%,1.67%.Experiment determined the total flavonoids in lemon, and the linear relationship is goodwhen flavonoids standard was concentration of0-25ug/mL, R2=0.9979. Determination is377.6ug/mL, precision and repeatability was good, RSD6.6%.Method of ultrasonic waves was used in the extraction of lemon limonin, and UV andHPLC were used in the analysis and detection, and limonin standard is good when theconcentration was40mg/L-80mg/L, R2=0.999. The limonin wasn’t detected in lemon fruitjuice. The content was61.04mg/L in lemon seed, repeatability and precisionwere good, RSDwere1.84%,1.93%.Lemon polypeptide was preliminary separated and detected, and the polypeptide whichwas5KD-30KD was centrifuged and catched. Result of Biuret Color Reaction was fromcolorless to lightblue, and finally purple, it showed it contained polypeptide or protein. Theresult of SDS-PAGE showed: the samples appeared a strip in the range of10kD-15kD, andthe strip gradually deepensed with the increase of sample size; The RT-HPLC was subjectedto elute the sample and recombinant human insulin standard, the Results showed that thesame absorption peaks apprared at almost the same retention time. The gradient elution conditions was selected, result showed: the flow rate of mobile phase was0.8mL/min,0-1min:A was90%, B: Liquid is10%;1-30min:A is40%, B is60%.At the same time, the experiment measured the content of Cu, Zn, Fe, Se. Different partsof the lemon were digested according to specific digestion procedure. The three elements ofCu, Zn, Fe, were determined by flame atomic absorption spectrometry. And the content ofCu, Zn, Fe in different parts were: Lemon juice:0.709μ g/kg,1.244μ g/kg,14.312μ g/kg.lemon peel of0.787μ g/kg,2.430μ g/kg,12.971μ g/kg Lemon seed:3.384μ g/kg,5.948μg/kg,29.773μ g/kg. RSD was0.44%-1.19%, the recovery rate was98.6%-108.3%, the RSDof repeated experiments in the determining of Cu, Zn,Fe were in1.31%-7.58%. Method ofAFS was used in the determining of Se, the linear was good when standard was1μ g/L-32μg/L (R2=0.9998), RSD was0.26%, the recovery rate was80.0%-112.5%. The content Se inthe different parts was: fruit juice of0.0030μ g/L,0.0005μ g/L lemon peel, seed0.0014μg/L.The RSM was used in the study on Rapid Propagation Technology. And the projectprobed the conditions, the concentration of phytohormone in the induction of callus androoting. The scheme optimization of culture:6-BA1.95mg/L, GA1.46mg/L, IBA0.97mg/L; NAA0.40mg/L, BA1.76mg/L, activated carbon2.12mg/L; time10.7h, temperature34.99℃, light1712Lx, humidity50%, humidity.
