According to the past three national oral epidemiological survey, caries today is still amajor oral disease to our residents and which coveres all ages. The situation never changed alittle in the past30years. In addition, the oral health of our people was not good from otherresearches of gingivitis and periodontitis. Plaque and gingivitis are common in all ages andalso there are no sign of being good.This thesis focuses on common oral problems and the need of tooth whitening, and withthe purpose to design and evaluate toothpaste which is effective in antibacterial, repair andwhitening. What to do as follows: Firstly the toothpaste proves to be inhibitive to pathogenicbacterium; Secondly, with the control of plaque, it can strengthen the ability of anti-caries byremineralzation on the decayed enamel; Finally, the toothpaste can polish the repaired enamelthat will make the surface of tooth smooth and gloss, which turn out to be whitening tooth.1Antibacterial test and evaluation1.1Diluting propolis and chrysanthemum extract with agar plate dilution method andfor cetyl pyridinium chloride using nutrient broth dilution method, threeantibacterial agents were diluted to the same concentration. Mix and culture eachdifferent concentration antibacterial liquid with medium and the4oral commonpathogenic bacterium together, measuring the size of inhibition ring to test theMinimal Inhibitory Concentration(MIC) of the three antibacterial agents. Result:The three antibacterial agents are inhibitive to pathogenic bacterium, the inhibitionability order is as follows: CPC>propolis extract>chrysanthemum extract.1.2By using Kirby-Bauer method with blank toothpaste as negative control, comparethe antibacterial ability in toothpaste (0.1%,0.3%,0.5%propolis extract;0.1%,0.3%,0.5%chrysanthemum extract;0.05%,0.1%cetyl pyridinium chloride).Result: The toothpaste with these three antibacterial agents are also inhibitive topathogenic bacterium, the inhibition ability order is as follows: CPC>propolisextract>chrysanthemum extract.2Enamel repair test and evaluationTake15days demineralization and remineralization cyclic test using bovine tooth,choose some qualified teeth as the experiment samples. Then compare5%HAPtoothpaste with0.14%sodium fluoride solution (Positive Control) and with water(Negative Control). Result: After15days demineralization and remineralization cyclic test treated by5%HAP toothpaste, the tightness and the remineralizationhardness of the repair layer on the tooth surface are better than another treatment.The hardness of the layer increases44.84±7.60%.3Whitening3.1RDA test: Irradiate the test tooth sample with standard neutron current, thephosphorus in the sample will quantificationally turn into radioactive32P, thendetect the seriflux after standard brushing test, the result is RDA value aftercalculation. Result: The dentin abrasion decreases with the treatment of2%perlitetoothpaste.3.2PCR test: Dye the tooth sample with standard method, take qualified ones to havestandard brushing testing, then detect the L value with colorimeter before and afterbrushing. The change of whiteness reflects the cleaning ability of the toothpaste.Result: The cleaning ability increases with the treatment of2%perlite toothpaste.3.3EPP test: Take pretreatment of the tooth sample and then the standard brushingtesting, the polish ability of the toothpaste will take out with gloss test before&after brushing. Result: The polish ability obviously increases with the treatment of2%perlite toothpaste.Above all, the antibacterial, repair and whitening ability of the toothpaste are proved within vitro test, which set up a good foundation for the following product research and forclinical test.
