Sugar refinery mud filter is one of the main waste in cane sugar mill, in which contain ascertain amount of nutrients. If we discard the mud directly, not only cause environmental pollution, waste the resources. At the present, doing with the mud filter and recycling are researched abroad concentrating mainly on extraction of nutrients and production of compound fertilizer, this research which about cultivate microorganisms to be rhizobium inoculants is less.In order to put the sugar refinery filter mud as recycling raw material, this article use various ways to determine the contents of components in the filter mud and the contents of metal elements. The research provide reliable data for developing and utilizing of filter mud, also make fully development and utilization of sugar refinery filter mud.Fresh sugar refinery filter mud was used as a medium for rhizobia tested, through determine OD600to analysis the growth conditions, rhizobia cultivated under the same conditions with YMA medium OD600contrast the test set optimization solution. There are four single-factor experiments:humidity control of filter mud is65.92%(the moisture content of fresh filter mud after filtering),72.97%,84.85%respectively; controlling factor of total sugar and total nitrogen ratio in filter mud is5:2,10:7,1:1,7:10,2:5; pH gradient is6.5,7.0,7.5,8.0,8.5,9.0; temperature gradient is24℃,28℃,32℃,36℃,40℃. Rhizobia with3,5-dinitrosalicylic acid method measured logarithmic phase when the medium at the end of the residual sugar side reflect the nodule bacteria growth.The results show that through control the environment, use filter mud in cane sugar mill directly to cultivate this experiment used fresh soybean rhizobia is feasible. Compared with best YMA rhizobia culture medium the industry, obtaining the growth of strains is similar to YMA medium in the same cultivation time by control the condition of four single-factor experiments. Through the response surface analysis method can predict the best technological conditions combination of the cultivation.This expriment reachs that the nitrogen content in the filter mud samples is3.63%, the content of crude protein is22.68%, total sugar content is about0.61g/mL, the content of phosphorus is1936.8μg/mL. Through the single-factor experiment and response surface method, analyze and optimize that the best growth condition of legume bacteria are:T=29℃, pH=7.5, the ratio of total sugar and nitrogen is7:10, humidity is72.97%, the size of inoculums is51.3mL/kg. Dehydration filter mud can be used as a carrier of the growth of rhizobia.
