In this paper, the horse blood as experimental material, the ultrasound technology, themicro-capsule technology and the pharmaceutical technology were used, to research on hemetablets was prepared by horse blood, establish quality standards and analyze the function of heme,to provide an experimental basis for production. The main results are shown below:(1) Through single factor test to determine the important factors for affecting the yield ofheme were ultrasonic power, ultrasonic time, extraction pH and deionized water content, throughresponse surface analysis and optimization of the extraction of the optimum conditions for horseblood heme that ultrasonic power was338W, ultrasonic time was10min, deionized water contentwas5times the size of red blood cells (RBC), and extraction pH was5.5, the yield of hemereached maximum0.42g/100mL.(2) Compared three wall material embedding rate of carrageenan, gelation, and gum arabic,showed that gum arabic was the best wall material. Based on the single factor experiment, throughresponse surface analysis determining the optimal parameters of microencapsulated heme thatpre-frozen time was16h, wall mass concentration was15g/100mL, wall material content was19times the size of core material. Under these conditions, the embedding rate was87.08%.(3) Studied on the wet granulation tableting technology was used to suppress horse bloodheme tablets, after parameter optimization, to determine the preparation of heme tablets was thatcorn starch, microcrystalline cellulose and microencapsulated heme powder uniformly mixed in aratio of1:1:8, over80mesh sieve,75%ethanol solution as a wetting agent for granulation, andover40mesh sieve, dried at50℃in30min, pack through40mesh sieve, added0.5%magnesiumstearate were mixed uniformly compressed. The heme tablets’ type were complete, surface weresmooth, the weigh of each tablet was about0.427g, the content of heme was about7.76mg, thecontent of Fe was about0.77mg. In addition, disintegration and microbial indicators were inaccord with Pharmacopoeia standards.(4) By fed with low iron diet combined with the establishment of SD rats tail bleeding modelof iron deficiency anemia, after then orally administered to rats continuously28d horse bloodheme, rats’ hair were luster, bright, stool dark brown and dilute. Body weight of made module rats were lower than normal body weight before administration, and the difference wassignificant(P<0.05), the weight of the rats in each dose group had no difference to compare tomodel group(P>0.05), to show that the heme had no significant effect for gaining weight of irondeficiency in rats. After administration comparing with model group, the horse heme of low,medium and high dose groups could significantly increase Hb and RBC and HCT content in irondeficiency, and difference was significant(P<0.05), at the same time, the three content can reachthe normal range. So the horse blood heme has a certain effect to improve the situation of irondeficiency.
