Tobacco is the most widely planted crop in the world. It not only can be made of cigarettes, cigars, pipe tobacco, hookah, snuff, chewing tobacco and other tobacco products, but also in terms of organic pesticides and medicine, has great value, such as tobacco whole plant can be used for pesticides, especially nicotine content; tobacco can also be medicinal, such as it can be used for anesthesia, sweating, sedation, emetic agent and so on.Whether from the number of smokers, tobacco production, or from the cigarette output and tobacco growing area, our country is ranked first in the world.Tobacco is an important source of income in state and local taxation. As the smoking and health problem dues more and more attention, improving the quality of tobacco and its products, and reducing harmful ingredients in tobacco have become the eternal theme of the development of China’s tobacco industry.The content of tobacco oil is low, the composition is complex, and it has a direct relationship with the quality of tobacco. But tobacco oil is a general concept, at present, there is no accurate definition about the concept of tobacco oil. In order to improve the quality of tobacco, the systematic research conducted about tobacco oil is necessary.The main work of the article:1.With the method of simultaneous distillation extraction as a means and methylene chloride as solvent, we systematically studied and optimized the extraction process of tobacco volatile oil by single factor and orthogonal experimental.The optimum conditions: the extraction time was 12 hours, the soaking time was overnight(12 hours), the concentration of sodium chloride was saturated(360g / l), the material liquid radio was 1:20, the particle size was 40 mesh. we detected and analyzed the volatile oil of tobacco by the methed of GC / MS,we detected 48 kinds of substances.2.With the method of ultrasound soxhlet extraction as a means and petroleum ether as solvent,we systematically studied and optimized the extraction process of tobacco oil by single factor and orthogonal experimental. The optimum conditions: the extraction time was 4 hours,the ultrasonic power was 70 watts, the ultrasonic time was 40 minutes, the soaking time was 12 hours, the material liquid radio was 1:8,the particle size was 40 mesh.We detected and analyzed tobacco samples coming from four different places such as Pingdingshan, Fujian, Luoyang and Zambia, we detected 97 kinds of substances. Compared with the volatile oil of tobacco, tobacco oil was mainly obtained by the volatile oil of tobacco and the lipid of high molecular weight,such as fatty acids, grease, lipids, sterols, terpenoids and so on.3.We extracted and analysed 28 reference samples of national flue-cured tobacco by the method of ultrasound soxhlet extraction. With the standard of extraction rate of tobacco oil, we studied the variation of tobacco oil in different factors, such as parts, colors, maturity, grades and so on.4.The peaks in 21.956 minutes(nicotine), 47.560 minutes(pinane), 57.557 minutes(palmitic acid) were selected as characteristic peak. To explore the way to improve tobacco oil, we analysed the tobacco samples of applying different base fertilizer from two perspectives of extraction rate of tobacco oil and abundance of characteristic peaks.
