The oil extract from tea seed is called Tea seed oil. The content of unsaturated fatty acid in Tea seed oil is up to 82%[1].Tea seed oil is rich in polyphenols, phytosterols, qualene, vitaminE and other active ingredients.《the people’s Republic of C hina Ministry of health in 2009 eighteenth announcement 》 in the formal approval of the tea seed oil as a new resource food [2].The oil extract from camellia seed is called O il tea camellia seed oil. The proportion and content of fatty acid in camellia oil is similar to olive oil[3], camellia oil can be comparable with the olive oil in high- grade health edible oil.《C hinese food structural reform and development progra》vigorously promote the camellia oil.Unsaponifiable matter refers that not reaction with NaOH in Saponification reaction. This thesis focused on the research of unsaponifiable matter in tea seed oil and oil tea camellia seed oil, explored the composition of tea seed oil and camellia oil unsaponifiable and found the method for separation and purification of unsaponifiable matter, and squalene in tea seed oil was determined by high performance liquid chromatography. To provide the experimental basis for the development and production of tea seed oil and oil tea camellia seed oil. The main research results are as follows:1.Determine the unsaponifiables content of tea seed oil and oil tea camellia seed oil.According to the national standard GB/T 5535.2-2008/ISO to extract of tea seed oil and oil tea camellia seed oil unsaponifiables and determine the content, measured in crude oil unsaponifiable matter content is 5.63%, oil tea camellia seed oil in crude oil unsaponifiable matter content of 5.56%. The microwave assisted extraction of extraction of tea seed oil and oil tea camellia seed oil, the extraction of tea seed oil and oil tea camellia seed oil unsaponifiab le matter content were 7.51%±0.51% and 7.36%±0.37%.2.Determine the components of tea seed oil and oil tea camellia seed oil unsaponifiables.Analysis the tea seed oil and oil tea camellia seed oil unsaponifiable components by GC-MS. The results showed that: 77 peaks were separated from tea seed oil unsaponifiables through GC-MS and identified 20 substances, including lanosterol content was the highest 5.91%, and alpha tocopherol, jasmone, squalene and other substances.Through the GC-MS from Camellia oleifera seed oil unsaponifiables in 56 peaks were separated, identified 13 material, where in the squalene content was the highest 2.69%, and geranyl linalool, tocopherol, ergosterol substance. Determined the content of total plant of tea seed oil and oil tea camellia seed oil phytosterol by phosphorus sulfur Tiefa, obtained the total content of the phytosterol tea seed oil was 2.07±0.001 g/kg, the total content of the phytosterol from oil tea camellia seed oil was 3.565±0.002 mg/kg.3.Separated the tea seed oil and oil tea camellia seed oil separating unsaponifiables to purification.The petroleum ether and ethyl ether as the agent, the volume ratio of 9:1, the tea seed oil and oil tea camellia seed oil unsaponifiables preliminary TLC separation, get six ribbons. By UV spectra showed the results of preliminary separation, determination of spectral scanning curve components between UV 200~350 nm. The results showed that: the tea seed oil unsaponifiable matter separated by thin layer of six bands in 200~350 nm waveform is different, and the maximum absorption peak is different, the maximum absorption peak wavelength were: 237, 242, 240, 233, 238 nm and 232 nm; oil tea camellia seed oil unsaponifiables through thin layer separating the six strip in the 200~350 nm waveform is different, and different maximum absorption peaks. The maximum absorption wavelength were: 236, 241, 240, 234, 238 nm and 232 nm. By thin layer chromatography, reached a preliminary separation of tea seed oil and oil tea camellia seed oil unsaponifiables component requirements.Using column chromatography purification of tea seed oil and oil tea camellia seed oil unsaponifiables. The results showed that: the content of hydrocarbons, three terpene alcohols, 4- methyl sterols and sterol of tea seed oil unsaponifiable matter were 15.75%, 54.9%, 1.5% and 12%, the content of hydrocarbons, three terpene alcohols, 4- methyl sterols and sterol of oil tea camellia seed oil unsaponifiable matter were 21%, 58.15%, 0.55% and 11%. The content of hydrocarbon oil tea camellia seed oil unsaponifiables and three terpene alcohols is higher than that of tea seed oil, but the content of tea seed oil unsaponifiables in 4- methyl sterols and sterol is higher than that of oil tea camellia seed oil.4.Determine the content of tea seed oil squalene. HPLC method for the quantitative analysis of tea seed oil squalene. The separation of oil samples in tea squalene by C18 column, the accurate calculation method of content of squalene standard used abroad. The results showed that: with methanol/acetonitrile(60+40, v/v) as mobile phase, detection wavelength at 210 nm, flow rate of 2 mL/min, column temperature of 30 DEGC, and can effectively separate the groups of tea seed oil in the sample, and then detected by HPLC of squa lene content of tea seed oil was 4.1 mg/kg. When the squalene quality concentration of 20~500 mg/L, the linear relationship between peak area and concentration of good, the linear correlation coefficient(R2) was 0.9990; the average recoveries for 84.2%~90.1%, the relative standard deviation(RSD) of less than 6.98%(n=4), determination limit of this method was 2.7(LOQ) mg/kg.
