Flavonoids,full of biological activities and medicinal value and widely exist in plants,are being researched and exploited by more and more people.Based on this,this article,on the one hand,has explored the extraction methods of flavonoids in Mimosa and finally determine the extracted parts.On the other hand,the purification and qualitative methods of flavonoids was established.This paper also tested eight kinds of sulfonamides in cosmetics by HPLC.This article is divided into four chapters,the research content is as follows:In chapter 1: Summarized the methods of extraction,separation and qualitative of flavonoids,and described the background,research purposes.In chapter 2: The extraction process of mimosa stem and leaf was optimized,and the extraction part was determined.The optimal extraction conditions of the total flavonoids in the stem and leaf of Mimosa were optimized.Considering the solvent composition,the ratio of the ingredients,the ultrasonic time,the ultrasonic temperature and the soaking time,the optimum conditions were selected by the single factor test,and then optimal extraction conditions were determined by orthogonal test.The content of total flavonoids in the stems and leaves of the mimosa leaves was compared under the optimum extraction conditions.The extract part was determined taking consideration of proportion of stem and leaf and root in the plants.In chapter 3: The separation,purification and qualitative methods of flavonoids in Mimosa were established.The flavonoids in sensitive plant were purified by Diaion HP-20 macroporous resin combined with Sephadex LH-20 gel resin.During the process,the collected components were monitored by analytical HPLC.Retention time qualitative method was used to qualitative coumarin and flax isoflavones.Yellow crystal purified by macroporous resin and gel resin was qualitative as avicularin by NMR.In chapter 4: Established a determination method of eight sulfonamides in cosmetics by HPLC.The optimal separation conditions were obtained by optimizing the condition parameters that affect the separation.In the range of 0.10~100 mg/L,8 kinds of sulfonamides had a good linear relationship.The correlation coefficient(r)was 0.9999,the limit of detection was 0.10~0.25 ?g/kg,the recoveries rate were between 87.1~128.7%.The relative standard deviation was between 4.6~8.6%(n=5).The method has high sensitivity,short analysis period and great reference value for the qualitative detection of sulfonamides.
