Scutellaria Baicalensis, the root of Lamiaceae Herbaceous Plants, is one of the commonly used Chinese herbal medicines for long history. Its main effective constituents are flavonoids compounds. Baicalin and Baicalein are the two with the highest percentage in all the elements. Scutellaria Baicalensis is with the significant biological activity, such as anti-inflammatory, immunoregulation, anti-allergic reaction, etc. In addition, it has been recorded that Scutellaria Baicalens is also with the efficacy for suppressing the melanin, anti-microbico. This study has explored the multiple efficiency and safety of Scutellaria Baicalensis used in cosmetic based on the modern biological theory, which has provided ideas and theoretical basis for the research and promotion of the multiple efficiency cosmetics with Chinese herbal medicine. The key results are shown as below:1) As the main effective components, Baicalin and Baicalein were extracted from the Scutellaria Baicalensis by high-performance liquid chromatography for qualitative and quantitative determination. They had been tested by tyrosinase inhibition method to determine the target composition of Scutellaria Baicalensis extraction condition. The results demonstrated that the inhibiting tyrosinase ratio of Baicalein obviously exceeded Baicalin’s, when the concentration of Baicalein reached to 2 mg/m L, its Inhibition rate could reach over 60%.2) Baicalein was used as the target composition of Scutellaria Baicalensis extraction to study the effect of Baicalein under different temperatures, liquid/material ratios and extraction times to determine all the factor levels. And three factors and three levels response surface analysis tests were set up to establish regression model and polynomial fitting, which were draw process prediction model and validated. The results demonstrated that the optimum condition of Scutellaria Baicalensis extract is 2 h at 80 ℃ with a liquid/material ratio of 20 m L/g. After the tests, it could be concluded that the real value is consistent with the predicted values.3) Refer to the current release of alternative test methods from the Organization for Economic Co-operation and Development of the European Union, the Bovine Corneal Opacity and Permeability Test, In Vitro 3T3 NRU Phototoxicity Test and RBC Hemolytic Test were adopted to evaluate the safety of Scutellaria Baicalensis extraction. The results demonstrated that IVIS values was calculated to be less than 3 in BCOP test, the results is mild and no eye irritation. From phototoxicity test, the EC50 of Scutellaria Baicalensis extraction was 86.151 mg/m L without daylight, on the contrary, the EC50 with daylight was 90.048 mg/m L. After the nontoxicity through Phototox software of the curve comparison operations, in the hemolytic test, the half hemolysis rate of Scutellaria Baicalensis extract was 0.0449 g/m L, which could be considered without irritation.4) The tyrosinase inhibition method, DPPH radical scavenging test, Hyaluronidase inhibition method, mice auricle swelling test, microbial method were used to study on multiple efficiency of scutellaria Baicalensis extraction. The results demonstrated that when Scutellaria Baicalensis extraction concentration was 0.2 g/m L, its inhibition rate against tyrosinase could reach 75.87%, half-inhibition concentration was 0.106 g/m L. When the concentration was 0.01 g/m L, its clearance rate against DPPH could reach 93.09%. In addition, concentration of Scutellaria Baicalensis extract was 0.1 g/m L, hyaluronidase inhibition rate could reach 72.1%, its half inhibitory concentration was 0.567 g/m L. Scutellaria Baicalensis extraction was also with a certain ability to inhibit the P.acnes, that results when the MIC was 30 mg/m L, its antibacterial efficiency was slightly higher than that of water-soluble tea tree oil.5) Combined with the skin feeling and stability evaluation, Scutellaria Baicalensis low-sensitive whitening emulsion was made by optimizing the emulsifier and thickener and the Scutellaria Baicalensis extract content based on the emulsion formulation, and had human skin patch test and periodic whitening effect evaluation. The results demonstrated that, the best emulsifier system was Tego Care 450 and enzymatically decomposed lecithin compounded, and the best thickener system was xanthan gum and Sepiplus 400 compounded. Scutellaria Baicalensis extract adding range was 0.2 ~ 1.5%, its effective optimum adding amount was 1.5%, after the use of 60 days it could make the skin melanin value lower about 25.
