Chrysanthemum is one of the ten famous flowers in our country, it has a high ornamental value, chrysanthemum also has high medicinal value and nutritional value. Now, study of chrysanthemum just focuses on volatile oil, choline and flavonoids at home and abroad. The study of polysaccharide from chrysanthemum is less. So, this research launched a series of studies of polysaccharide from chrysanthemum, the extraction, separation and purification of polysaccharide from chrysanthemum, in vitro antioxidant activity and antitumor activity of polysaccharide from chrysanthemum are contained in this research.The extraction technology of CMP were investigated and extraction process was optimized. There are three extraction process: water extraction, ultrasonic assisted extraction and enzyme extraction method. All of the extraction method were investingatated. From single factor experimental the best extraction condition of the water exraction was that extracting time, material liquid ratio and extraction temperature were 2h, 25 m L/g, 90 ℃. In these conditions chrysanthemum polysaccharide rate was 11.74%. The best extraction condition of the ultrasonic assisted extraction was that extracting time, material liquid ratio, extraction temperature and ultrasonic power were 35 min, 25 m L/g, 70 ℃ and 60 W. Under the condition the chrysanthemum polysaccharide yield is 12.68%. The best extraction condition of the cellulose hydrolysis extraction was that extracting time, material liquid ratio, extraction temperature and enzymatic volume accounted were 2 h, 40 m L/g, 2%. Under the condition the chrysanthemum polysaccharide yield is 13.66%.On the basis of single factor, the response surface analysis method was used to analysis the influence of water extraction on extract the polysaccharide from chrysanthemum. Adopting the three factors and three levels, according to the experimental plan to experiment, the experimental results that the best technological conditions of extracting time, extraction temperature and solid-liquid ratio were 2.51 h, 91 ℃,24.5 m L/g, and under the conditions the yield of polysaccharide chrysanthemum reach 11.79%.CMP was made after polysaccharide extracted from chrysanthemum in the optimal process conditions of water extraction. Then using the resin and hydrogen peroxide methods to decoloring, Sevag method to remove protein and then ethanol subsided. Respectively with intercept relative molecular mass of 300 k Da and 100 k Da two ultrafiltration membrane ultrafiltration of CMP. CMP was divided into three kinds of polysaccharide components, the CMP-1 which relative molecular mass is more than 300 k Da, the CMP-2 which relative molecular mass between 300 k Da and 100 k Da, the CMP-3 which relative molecular mass is less than 100 k Da. Besides, this research was study on the influences of the membrane flux of ultrafiltration, the aspects includes out of the inside and outside differential pressure of ultrafiltration membrane, samples of solution temperature and solution concentration. Get the most suitable experimental conditions of ultrafiltration: membrane inside and outside differential pressure is 0.03 MPa, solution temperature of is 40 ℃, solution concentration of is 1 mg/m L.To study antioxidant activity via DPPH·, ??2-·,and ·OH free radical scavenging and reducing power, the results indicated polysaccharide from chrysanthemum had good ability of scavenging free radicals and strong dose dependent. Antitumor activity of polysaccharides from chrysanthemum was determined on two kinds of cancer cells human hepatoma Hep G-2 cells abladder cancer and human breast cancer MCF-7 cells by MTT assay. The results showed that CMP, CMP-1, CMP-2, CMP-3 had inhibitory effects against human hepatpma Hep G-2 cells. And all polysaccharides had low inhibitory effect on human breast cancer MCF-7 cells.
