Odoratum is a dry rhizome of Liliaceae, which has a broad prospect of development and application. With the development of studies, Polygonatum polysaccharide (POPS) is paid attention to by the domestic and foreign scholars. In this paper, the separation, structure, activity and application of Polygonatum polysaccharide were studied.In this study, two extraction methods of POPS, conventional water extraction and microwave method. Based on the single factor experiment, the optimum parameters and scopes were ensured by the method of orthogonal design. The result showed that the optimum parameters of water extraction were80℃,2.5h and the ratio of material to water was1:30, extraction times were2times, the extraction rate of polysaccharide was up to8.69%; The optimum parameters of microwave extraction method were100w、4min, the ratio of material to water was1:20, extraction times was1time, the extraction rate of polysaccharide was up to11.95%. The crude polysaccharide was attained through deproteination by sevage method, being precipitated with alcohol and being lyophilized. Polygonatum polysaccharide is divided into two components POPS-A and POPS-B by using dialysis bags. POPS-B1and POPS-B2were isolated from POPS-B through Sephadex G-75.The purity of POPS-B1was analyzed by optical rotations, chromatography with Sephedex G-25and UV absorption spectrum, indicating that POPS-B1was a homogeneous polysaccharide; The weight average molecular weight of POPS-B1was2800Da determined by HPLC; The FTIR spectrum showed that POPS-B1have-OH、C=O、pyranose ring、and mannose. The thin-layer chromatogram and GC was used to analyze their monosaccharide components and showed that POPS-B1was two kinds of monosaccharide including mannose and glucose with molar ratio of them as2.32:1.The results showed that POPS contained phenolic substances by researching on color reaction of Polygonatum polysaccharide with Fecl3. The antioxidant effects of different Polygonatum polysaccharide (POPS-A, POPS-B) were studied in vitro by different testing model. The results showed that POPS-A and POPS-B had certain antioxidant activities. The correlativity of their effects and the dose are positive, in a certain concentration range; Results of experiments on antibacterial and antifungal activities showed that POPS had some inhibitory effect on Escherichia coli, POPS-A is still no inhibitory effect on bacterial and fungal and POPS-B had strong inhibitory activity against Bacillus subtilis and Aspergillus niger.Polygonatum polysaccharide has significant hypoglycemic effect, regarding the polygonatum polysaccharide、bitter gourd and red beans as materials, the formula of the functional beverage of polygonatum was studied by orthogonal test. The optimum formula for the solid beverage of polygonatum was that bitter gourd powder, redbean powder and Polygonatum polysaccharide ratio1:8:0.5; The ratio of the β-cyclodextrin、oligomaltose and citric acid was0.2:2.2:0.01; The optimal dosages were0.11%CMC,0.09%xanthan gum and0.2%pectin.
