| ABSTRACT Herba elpmedii ,as traditional Chinese medicine, has been widely used in clinic. The leaves of Herba elpmedii were generally taken as medicine with benefiting kidney. In these years, much work has been taken to the leaves, but little to the roots. The flavonoids were considered to be the main active parts and the icariin as the effective component of the leaves.Firstly, in this paper other active parts of the leaves and the roots of Herba elpmedii were screened by using the vascular cell membrane chromatography (CMC) model. It was found that the ether extract of the roots of Herba elpmedii was active for the vascular cell. Secondly, the ether extract was separated by thin layer chromatography(TLC),column chromatography(CC) and high performance liquid chromatography(HPLC). Each component of the ether extract obtained by the separating methods was screened by using the vascular CMC model. The YYH-102705 and YYH-110805 were found to have the retention characteristics on the model. Then, both YYH-102705 and YYH-110805 showed that they can inhibit the contraction produced by norepinephrine on the rabbit vascular in vitro. So that, the screening results obtained by the CMC were found to have a good correlation with their pharmacological effects in vitro. The YYH-110805 and YYH-102705 were the effective components in the roots of the Herba elpmedii.Thirdly, the TLC and RP-HPLC were used for the comparative analysis of the effective components in the roots and leaves of Herba elpmedii. The results also showed that there were not the same effective components in the leaves of Herba elpmedii under the chromatographic conditions. In addition, the YYH-102705 and YYH-110805 were further separated and purified. Two yellow crystals were obtained. By the elements analysis, UV, IR, 1H-NMR and 13C-NMR, YYH-102705 has been identified as baohuoside-â…¡ and YYH-110805 as baohuoside-â… .In this paper, it was firstly studied on the difference and pharmacological action of the effective components in the roots and leaves of Herba elpmedii by using the CMC model at the levels of molecular and cell, and the baohuoside-â… and baohuoside-â…¡ were found to be two vasodilatation effective components. The results will provide scientific basis for exploiting, utilizing, qualitative and quantitative analyzing the roots of Herba elpmedii, and it was another successful example in the application of the CMC model.
