I studied the taxonomic status of Herba artemisiae subdigitatae and Herba artemisiae hedinii by the method of morphological taxono-my.I looked for the anatomical features of Herba artemisiae subdigitatae and Herba artemisi-ae hedinii with anatomical method.I researched the major ingredients of Herba artemisiae subdigitatae and Herba artemisiae hedinii by TLC\HPLC.I established a stability analysis method. I extracted the chemical ingredients-coumarin related to the medicinal efficacy using supercritical CO2fluid extraction method.In this paper,I identified Herba artemisiae hedinii with Herba artemisiae subdigitatae by literatures and source identification.I identified them on their macrostate,micromorphology and powder characteristics of leaf and stems.Herba artemisiae hedinii are bipinnatifid and pectination on morphological character.Herba artemisiae subdigitatae are feathered deep cleft on morphological character. Herba artemisiae subdigitatae has obvious secretory cavity,but Herba artemisiae hedinii is hard to find on the anatomical characteristics. They all have scopoletin.I had an investigation on thin layer conditions.It is concluded that the scopoletin could separate from other components with the temperature in the range of4℃to40℃,distance in8cm to15cm, the humidity in the range of35%~75%, treating ethyl acetate-methanol-concentrated ammmonia solution(17:2:1) as developing agent. They have differences in the number of spots on the TLC plate.I determined the the moisture content,ash content and extraction content in the herba artemisiae hedinii and herba artemisiae subdigitatae.The contents of moisture in herba artemisiae subdigitatae are between6.34%and6.44%. The contents of moisture in herba artemisiae hedinii are between5.86%and6.23%. The contents of total ash in herba artemisiae subdigitatae are between7.38%and7.53%. The contents of total ash in herba artemisiae hedinii are between5.71%and7.73%.The exacts in herba artemisiae subdigitatae are between8.19%and10.12%. The exacts in herba artemisiae hedinii are between25.74%and27.94%.I determined the content of scopoletin in the Herba artemisiae hedinii and herba artemisiae subdigitatae by high performance liquid chromatography (HPLC). The chromatographic conditions are optimized. Chromatographic columns of them are Agilent TC-C18column (4.6mmx250mm,5um).Column temperatures are30℃and35℃. Mobile phases are methanol and0.3%phosphoric acid solution. The flow rates are1.0mL/min and0.8mL/min. Herba artemisiae hedinii is with gradient elution. Herba artemisiae subdigitatae with isocratic elution.The detection wavelengthes are344nm.At the same time,I had an investigation ofmethodology. It is concluded that the mass concentration of scopoletin and peak area had good linear relationship within the range of measurement of0.004~0.03mg/ml and0.005-0.025mg/ml.The regress equations are y=6.587E+07x+9863.3and y=1.766E+07x-1304.2467.The average sample recovery rates are100.64% and99.75%.This method is sensitive and accurate,simple and rapid.I determined the contents of scopoletin in9group of Herba artemisiae hedinii and2group of herba artemisiae subdigitatae Using this method.It is result that the contents of scopoletin in Herba artemisiae hedinii are0.6811~0.8549mg/g. The contents of scopoletin in herba artemisiae subdigitatae are0.3813~0.6146mg/g. The contents of scopoletin in Herba artemisiae hedinii are higher than herba artemisiae subdigitatae.It can be the index marker of their quality control.They are different in medicinal properties,anatomy. Although they all have scopoletin,their contents are different. They should not mix with each other.They should establish the quality standard separately. In this paper, the preliminary draft of their quality standards were drawn up. It is proposed preliminarily that the moisture content of the herba artemisiae hedinii and herba artemisiae subdigitatae should be lower than6.5%.The total ash content of the herba artemisiae hedinii and herba artemisiae subdigitatae may not be over8.0%. The content of extract in the herba artemisiae hedinii shall not be less than25.0%calculated on dry goods. The content of extract in the herba artemisiae subdigitatae shall not be less than8.0%calculated on dry goods. The contents of scopoletin in Herba artemisiae hedinii should not less than0.6mg/g.The contents of scopoletin in herba artemisiae subdigitatae should not less than0.3mg/g.Treating scopoletin as reference substance,I established the ultraviolet spectrophotometric method of determination of total coumarin content in Herba artemisiae hedinii.The average recovery was99.02%,RSD1.25%,the results show that the method is rapid, simple,sensitive and accurate.The contents of total coumarin in Herba artemisiae hedinii are between1.205%~2.340%using the method. This method not only as evaluation index of total coumarin in Herba artemisiae hedinii in the extraction.At the same time,it provides referential experience and theoretical basis for determination of total coumarin content in other medicinal plants. Extraction index is coumarin content.The process conditions of coumarin in the Herba artemisiae hedinii are discussed by Supercritical CO2extraction technology. The factors of affecting extraction effect have carried on the comprehensive investigation.The extraction conditions were optimized through orthogonal experiment.The modifier is95% ethanol.Its dosage and mass ratio is3:1.Extraction pressure is25MPa. The temperature is45℃. CO2flow rate is20L/h..Extraction time is2h.The extraction rate is87.3%.
