In this paper, the extraction, separation, purification and analysis method of Bio-active composition Aucubin(AU) were investigated in the Chinese medicine Eucommia ulmoide oliver. The research is composed of four main parts:(1) Extraction prccess of AU with organic solution was studied on detail. The optimum parameters of extraction is gained : Which is 72% EtOH-H2O, temperature of extraction is 65C , time of extraction is 60 min, the ratio of solid to liquid is 1:12 and the extraction ratio of AU can overpass 85%, when the times of extraction is more than three.(2) The purification process was completed by macroteticular resin. AU was absorbed well by S-8 resin and eluted by 50-60% EtOH.(3) The research on spectrophotictric determination of AU is carried out. New analysis method is that: In the solution of 1.0mL AU, 2.2mL EtOH, 1.6mL E-reagent and 0.5mL acetic acid are added, then heated up for 12 min at 66 C and cooled for 10 min and the absorption is detected in the 594nm wavelength.(4) A Rp-HPLC method is establish for simultaneous determination of geniposidic acid, chlorogenic acid and geniposide in Eucommia, The effect of Meoh concent, the acidic additive and pH were investigatedon the separability of compounds. The optimum condition of chromatography is obtained with the flow phrase MeOH-H2O-H3PO4 (12:87.96:0.04) and tge flow speed is 1.0mL/min. The capacity factors of three compounds range from 3 to 13, the peaks of three compounds can be separated in the base line and analysis time was in 30min, and the retention times of geniposidic acid, chlorogenic acid an geniposide were 6.7min, 10.5min, 21min respectively.
