The extraction, analysis and purification method of vinblastine were reviewed. A improved technics of vinblastine purification through column chromatography was studied. The extraction of vinblastine from Catharanthus was also studied in the paper.At first, the HPLC method of analysis was established. ODS column ( φ 4.6mm X 25cm) . Mobile phase ethylenediamine- NANOpure ultrapure water-methanol 5:295:700, pH=7, velocity 0.5ml/min, UV267.The content of vinblatine in Catharanthus were determined as 4.57/10000. The extraction effect was ascended when the solvent polarity ascended, methanol attend by vitriol was the best solvent selected. Tempratrue raised was propitious to extraction, but the product was destroyed when tempratrue higher than 50℃, 40℃ was good.There were two steps for purifying vinblastine to purity 96%, first through alumina column chromatography, second through silica gel column chromatography. It could remove front impurity through alumina column, back impurity through silica gel. At the first step, chloroform-hexane 7:3 was the best mobile phase selected. Thus, if vinblastine/station phase(w/w) was 0.035 , the product purity was improved to 79%, and the recovery was about 95%.In the second step silica gel column, selection of the mobile phase was guided by TLC chromatography. Then mobile phase system was selected at silica gel column, chloroform-methanol-hexane (9:1:5) was the best mobile phase selected. Then, the product purity was 94,4% and recovery was 80%. After crystaled, the last product purity was 96%. The whole recovery was 48%, There were 15 percent higher than traditional technics.At last, A mathematical model was established with the assumptions of linear equilibrium and no mass transfer resistance inside the solid particles, which can describes the linear chromatography process.
