Based on AGILENT 6890 GC, a preparative GC system (PGC) was designed. A 16-position SD valve was the key part for the collector of the PGC. By setting preparative cycle on Chemistation's time event window,16 effluents could be collected one by on for once GC running. The collection part can be a cigarette, and the flavoring effluent will be automatically added onto it after GC separation. Conduits and 16-position SD valve, which were coupled to sources of thermal energy to prevent condensation within them, were all inert. The preparative experiments showed the purity of the collected fatty acid methyl ester was better than 98% for the well resolved peaks.Separation of four primary alkaloids in tobacco was carried out by using PGC. The purity of nicotine, nornicotine, anabasine and anatabine was 100,90.3%,81.8% and 93.3% respectively. Nicotine, nornicotine, anabasine and anatabine were identified by GC-MS and standards. The relation of four primary alkaloids and sensory quality of tobacco was studied.By the aid of automated top flavor adding function, the influence of several volatile and semi-volatile tobacco flavors on quality of tobacco was also studied. The results of this study indicated that some components will increase the aroma of cigarette, but the effects were varied.
