| Three kinds of polysaccharides were obtained from the Pueraria locata, Rehmannia glutinosa and Copis chinensis by water-extracting, alcohol-precipitating and Sephadex G-150 column chromatrography-separating. The results showed that they were homogeneous polysaccharides. Simotaneously, the molecular weight were obstained by Sephadex G-200 column chromatrography. The compositions of their single sugar were measured by HPLC. The basic structure was analyzed by HPLC, IR, periodate oxidation and Smith degeneration.Mw(PRC-l)=12.2 X 104, Mw(PRC-2)= 2.5 X 104, Mw(PRC-3)=7.62 X 103; PRC-1 consists of L-Rhamnose(35%), D-(+)-Xylose(l 1.5%), L-(+)-Arabinose(l 1.5%), D-Glucose(35%),D-Galactose(7%); PRC-2 concludes L-Rhamnose(5.3%), D-(+)-Xylose(2.8%), D-Fructose(16.3%), D-Galactose(1.7%), D-Glucose(73.9%); there are five kinds of single sugar including L-Rhamnose(12.4%), D-(+)-Xylose(16%), L-(+)-Arabinose(11.3%), D-Fructose(43%), D-Glucose(17.3%) in PRC-3. The data of IR spectrum revealed that PRC-1 is linked by α - glycosidic bonds and β -glycosidic bonds, PRC-2 and PRC-3 are linked by α - glycosidic bonds, while the backbone of PRC-1 is mainly linked by 1-6 and 1-2 glycosidic bonds, with a few 1-3 linkage, the backbone of PRC-2 is mainly linked by 1-2 glycosidic bonds, with a few 1-3 and 1-4 linkage, the backbone of PRC-3 is mainly linked by 1-6 glycosidic bonds, according to the results of periodate oxidation and Smith degradation. |
