Radix Codonopsis is Campanulaceae branch Codonopsis plants.The dry root is put into utilization,which may restore vital energy and raise the blood,enhance immunological function of organism.It is used to yin asthenia,breath weak and deficiency of vital energy and so on.The modern Pharmacology research confirms that Codonopsis pilosula has the biologic activity of anti-tumor,the enhancement immunity function and the anti-oxidation and so on,its main active constituent for codonopsis pilosula is polysaccharide in it.In this papers,we decide the most optimal parameter during all extracting process extracted,We studied the pharmacological action preliminarily and separated and purified the Lu radix codonopsis polysaccharides and obtained three homogeneous polysaccharides.The main contents of the papers are as follows:1.The optimization of extraction method of the radix codonopsis polysaccharides.According to the characteristic that monosaccharides dissolve to the water facilely,dissolve to the certain density ethyl alcohol also;but the polysaccharides dissolve to the water facilely,do not dissolve to the certain density ethyl alcohol.Monosaccharides,oligosaccharides and other micromolecule matters were removed by ethanol,this experiment used the method of water extraction and ethyl alcohol precipitation,and the experiment purpose is to obtain the techonology parameter by taking the polysaccharide from the Lu radix codonopsis take the extraction rate as detection index,at the same time analysed the primary and secondary factor with the orthogonal design method,screens the best extraction technology of the radix codonopsis polysaccharide. Considering the extraction efficiency and cost,the final optimization extracting method was that ten times amount water was put into the radix codonopsis decoction pieces,which was heated for one hours,the extracting solution was concentrated to 1mg/ml,and put into ethyl alcohol to precipitate when the density is 80%,the extracting rate of the polysaccharide is 26.45%.And 3 times of parallel experiments finally confirmed:The method is simple,stable and accurate,may be used in extraction of the radix codonopsis polysaccharide.2.The study of pharmacological action of the radix codonopsis polysaccharides.We studied the anticancer action in vivo with abdominal cavity S180tumor bearing mice,which was treated by gavage with radix codonopsis polysaccharides 1 g/kg·d,measured the aveage survival time after 7days,which is 34.29%.The results are as follows:radix codonopsis polysaccharides can lengthen survival time of abdominal cavity S180ascites tumor bearing mice. 3.The separation and purification of radix codonopsis polysaccharides.First,the crude radix codonopsis polysaccharides was separated and purified by DEAE-cellulose column,water and different density sodium chloride solution gradient elution,phenol- sulfuric acid method was used to detect various fraction,then every fraction was collected,vacuum drying again,and five polysaccharides subfractions were got:CPS-1,CPS-2(it was not studied further because of content fewer),CPS-3,CPS-4 and CPS-5.4.The study of the structure of the fine purification radix codonopsis polysaccharides.UV spectrum and high performance gel permeation chromatography tested purity of polysaccharides subfractions.The results are as follows:protein and nucleinic acid were not contained in all of them.The high performance gel permeation chromatography showed the polysaccharides subfractions were homogeneous polysaccharides.The molecular weight of CPS-1,CPS-3 and CPS-4 determined by high performance Gel Permeation Chromatography were 2500 Dal,1.15×105 Dal and more than 2000000Dal.UV spectrum tested the contents of four polysaccharides subfractions determined by sulfuric acid-phenol colorimetric and carbazole colorimetric method.The results are as follows:CPS-1 was neutral polysaccharide,the content is 76.87%by glucose.CPS-3 was acidic polysaccharide, the content of neutral polysaccharide and acidic polysaccharide is 28.51%and 59.63%by glucose;CPS-4 was acidic polysaccharide,the content of neutral polysaccharide and acidic polysaccharide is 25.02%and 50.68%by glucose.TLC showed that CPS-1 was composed of D-glucose,L-rhamnose.CPS-3 was composed of D-galactose,D-arabinose,glucuronate,possibly xylose in it.CPS-4 was composed of D-galactose,D-arabinose,galacturonic acid,possibly D-xylose and glucuronate in it.
