Traditional Chinese medicine are very important for Chinese to prevent and cure disease all along. The extraction is the modern types of TCM, it has wide international market space, so the industrialization extraction has a vast development prospect. The extraction, separation and purification are the key basic technologies of Chinese drug modernization and play a very important role in the GAP, GLAD, GDP, GME Therefore, the extraction, isolation and identification of the active components in TCM are more and more important for the modernization of TCM.The high-pressure homogenization extraction (HPHE) is a new way for realizing the modernization of Chinese drug. The technology is used to extract the essential components of herbs in this paper. Several key technical problems oh the technology are studied as follows:1. HPHE is use to extract the essential components. This is a new exploration and probing to extract the essential components from herbs. The effect factors of HPHE are analyzed with Hawthorn leaves in detail. The massive preparations experimental study has been conducted firstly and the massive repeated test are passed through by the foundation, then the basic parameters' ranges of HPHE to extract the Hawthorn leaves flavonoid are got as following: pressure 60MPa, holding pressure time 30min. These results not only provide experimental and theoretical basis more for extracting the essential components of epimedium leaf by HPHE, but also good experience for extracting the other different kinds of herbs and parts of a plant (including root, stem, leaf, flower and fruit).2. The process parameters of HPHE for the Hawthorn leaves are optimized by using the trial optimization design way, the mathematic model is set up, and the optimum process parameters are as following: pressure 60MPa, holding pressure time 30min, extraction solvent was ethanol of 70%(V/V), material/solvent (w/w)ratio was 1: 30(w/w). In this condition, the yield of flavonoids were up to 8.39%.3. The extracting method, process, characteristics and principles among HPHE, heat-reflux, and ultrasonic technologies are compared and analyzed systematically, which comes to the following conclusion. HPHE has the highest achievable yield for the essential components extraction of Hawthorn theaves on the condition of the optimum extracted solution. The achievable yield of HPHE is 1.08 times of reflux, 1.14 times of ultrasonic technologies, proved by the liquid chromatogram. The extraction time of HPHE is the least, only 1/8 of the reflux and 3/4 of the ultrasonic technologies. HPHE can be operated at normal temperature, without heating. It shows that HPHE is not only feasible and has a good development prospect, but also has the characteristics of operating simple, safe and the results in efficiency and time are better than that of the heat-reflux, and ultrasonic technologies ways.4. It is the first time to put forward the leaching mechanism of the essential components. The principle of HPET is not only explains the phenomena of achievable yield being high and time being short, but also provides some valuable references for the studies of the other extracting way.5. The osmosis resistance of the essential components in herbs is analyzed and the dynamic model of HPET is obtained.Generally, a new way, which is feasible to extract the essential components of the herbs without heating, is found out and the optimum process parameters is also decided. HPHE has the following good characteristics. 1) HPHE can be operated at normal temperature without the disadvantage of denaturalization of heating. 2) Solubility and transferring speed of most of the organic substances increase with the pressure increasing, in case, the achievable yield is high and the time of extraction is short. 3) The process parameters can be controlled and adjusted easily and cleaned easily for the HPHE equipment, a set of equipment can be used for different solvent and raw materials, and can carry out the extraction of different essential components. 4) HPHE is safe and economical.
