The Multi-stage Countercurrent Extraction (MCCE) technics of Traditional Chinese Medicine (TCM) such as Salvia Miltiorrhiza Bge, Notoginseng and Breviscapine were investigated. And the mechanism of MCCE process was preliminarily discussed. The aim of this study was to systematically clarify the applicability and superiority of MCCE in the field of TCM. The main works can be summarized as follows:1. The Multi-stage Countercurrent Extraction of Danshensu from Salvia Miltiorrhiza Bge was studied. The processing parameters were optimized using regression analysis and principal components analysis (PCA). The optimal condition of Danshensu extracted from Salvia Miltiorrhiza Bge was 1 hour extraction time, solvent to sample ratio of 12 mLg"1, 80 ℃, 2040 mesh of particle size. Compared with the refluence process, the MCCE process was 19.4 % higher in recovery, 75 % less in solvent consumption and shorter extraction time.2. The Multi-stage Countercurrent Extraction of notoginsenoside R1, ginsenoside Rg1, panax notoginseng saponins (PNS) from Notoginseng was studied. The multi-index optimization and regression analysis were used to optimize the parameters. The optimal process condition of notoginseng extraction was 70 % ethanol, solvent to sample ratio of 8 mL.g-1, 30 min, 50 ℃. Compared with the other processes, the MCCE had the advantages of lower temperature, 50 %70 % less solvent consumption and could deal with more herbs in unit time.3. The pilot-scale Multi-stage Countercurrent Extraction of Scutellarein from Breviscapine was studied. The optimal condition was 70 % ethanol, solvent to sample ratio of 10 mLg'1, 35 min extraction time and 80 ℃ extraction temperature. Compared with the other extraction process, the MCCE process had the advantages of higher efficiency, lower extraction temperature and less solvent consumption.4. The mechanism of Multi-stage Countercurrent Extraction was preliminarily dicussed. Take extraction of Danshensu from Salvia Miltiorrhiza Bge for example. The Danshensu extraction model was established.It was proved that the Multi-stage Countercurrent Extraction had systematical and wide applicability and superiority in the TCM field. It has the advantages of higher efficiency, lower extraction temperature and less solvent consumption. It can be widely used in the pharmaceutical industry.
