Wuweixiaodu decoction, Chinese herbal, ever listed in《Golden Mirror of Medicine》includes honeysuckle flower, wild chrysanthemum, dandelion, Tokyo violet herb and semiaquilegiae radix. Modern pharmacology indicates that Wuweixiaodu decoction has curative effects on anti-inflammatory, heat-clearing and strengthening immunization. It has been generally applied to the treatment of infectious diseases which were caused by burn infection and humid heat. Based on requirement of market and clinic medicine, using the extractum of Wuweixiaodu decoction we produced dispersible tablets and drop pills. These preparation are solid and can be taken orally. Seeking way in order that it can quickly exert action, reduce dose and enhance bioavailability. So the main research content is the extractive technique, treating process, the process and evaluation of preparation. This research includes: (1) the establishment of analytical methods of typical principles (2) the optimization on extraction and purification process of the active constituents (3) the pharmacodynamic study of Wuweixiaodu decoction (4) the preparation method of dispersible tablets (5) the preparation method of drop pills.According to literatures and experiments in this paper, TLC method was used to identify honeysuckle flower, wild chrysanthemum and Tokyo violet herb. Chlorogenic acid and caffeic acid were used as typical principles to determine the content of Wuweixiaodu decoction. HPLC methods were developed to determine their content in the herbals and the preparations. All of these methods were accurate and sensitive.The extraction process of the active constituents was studied by the criterion of content of chlorogenic acid and caffeic acid and the Dry-extracts content. Based on the results of single-factor tests, Orthogonal tests were used to optimize the decoction time, decoction times and solvent volume. Alcohol precipitation method, chitosan flocculating method and gel flocculating method were used to optimize purification process and the results indicates that the alcohol precipitation method is better than the others because it can make the solution precipitate fast and had higher transference rate of typical principles. The optimum method of extraction and purification process has a good repeatability.The pharmacodynamics experimentation of extract of Wuweixiaodu decoction which was gotten by using the optimal extraction and purification process indicated that Wuweixiaodu decoction had definite treatment on anti-inflammatory and heat-clearing.Based on the results of single-factor tests about the influence factors on disintegration time of dispersible tablets, the amounts range of wuweixiaodu decoction and excipients were set .The formulation was optimized on the orthogonal design by evaluating disintegration time and dissolution percents in 15 min. wuweixiaoduyin dispersible tablets were prepared according to the best components and preparing condition. The repeatability of three batch samples and stability of room temperature test showed that wuweixiaoduyin dispersible tablets were stable, well quality-controlled and met the purpose and requirement of dosage form design.To optimize the preparation process of wuweixiaoduyin drop pills by orthogonal design. Using the kind of base, ratio between drug and base, and the kind of cooler as affecting factors, the orthogonal test was conducted, to assess the formulation factors good or bad in the dissolution percent and appearance two indexes. Using the temperature of drug solution, temperature of cooler, drops distance and rate as affecting factors, the orthogonal test was conducted, to assess the preparation process good or bad in the appearance quality and the weight deviation of the dropping pills two indexes. The repeatability of three batch samples and stability of accelerating and room temperature test showed that wuweixiaoduyin drop pills were stable, well quality-controlled and met the purpose and requirement of dosage form design.
