Fibrauretinum has pharmacological action,such as:broad-spectrum antimicrobial,anti-virus,anti-inflammatory,increased-phagocytosis.It is a raw medicine used to prepare fibrauretinum Preparation,such as Tablets and Injection.The paper studies the process of fibraureain extraction and purification from Caulis fibraureae,Enriched alkaloids of Caulis fibraureae by macroporous adsorption resin and then purified of fibraureain and jatrorrhizine hydrochloride by chromatography is the first time.This purification process enhances both purification and rate,and Prepares jatrorrhizine hydrochloride.It laid the foundation for the comprehensive utilization of resources.The study and the experimental results are as follows:1.Determination of the alkaloids in Caulis fibraureaeAnalyzing the alkaloids in Caulis fibraureae by HPLC,palmatine hydrochloride and jatrorrhizine hydrochloride are the main ingredients;Determining the content of palmatine hydrochloride and jatrorrhizine hydrochloride.2.Study on Extraction Process of fibraureain by acid waterUsing palmatine hydrochloride content as a marke,The optimum extraction process is selected with cold dip;using single factor experiment method to examine the influence of such factors:the extraction time,frequency,solvent volume,sulfuric acid concentration.3.Comparative Study on purification process of fibraureain3.1 Optimizing process parameters on salting-out method and crystallization methodthe traditional technology of fibraureain is using salting-out method and crystallization method,Detailed technical parameters not been reported.In the study, the salting-out method and crystallization method of the key technical parameters were optimized.Under the optimum conditions,the fibraureain product could be obtained with purity of 94%,the yield is 0.62%.3.2 Purification process using macroporous adsorption resin and chromatographyMacroporous adsorption resin firstly,selected the patterns of macroporous resin;secondly,studied the adsorption capability of macroporous resin.Thirdly, studied the elution process of macroporous resin.The optimum purification process is as follows:D₁₀₁ macroporous adsorption resin,eluted with 6BVdistilled water,40% ethanol in turn,The purified alkaloids was collected in 40%ethanol eluting solvent. Under the optimum conditions,the crude product of fibraureain and the extract of jatrorrhizine could be obtained.Chromatography Using alumina column,the crude product of fibraureain is purified frome 82%to more 98.0%;Using silica gel,the extract of jatrorrhizine is purified of more 80%.using macroporous adsorption resin and chromatography,the fibraureain yield is 1.0%.
