Because of its higher efficiency, quicker analytic speed, lower consumption of mobile phase and less environmental pollution, Microcolumn Liquid Chromatography (micro-LC orμ-LC) is always used in the field of biochemical analysis, alimental and environmental analysis, nerve science, etc.Theμ-LC system was constructed via improving the conventional-type analytical equipment. Stability, as well as sensitivity of theμ-LC system was good enough to meet the quantitative and qualitative analysis. The retention time RSD and the peak area RSD of 7 times analysis were less than 0.14% and 1.1% when sample of Elite Corporation was used. Based on the gradient elution of theμ-LC ,seven compounds were separated within fifteen minutes. Compared with 4.6mmI.D.analytic column, 1.0mmI.D. column was much faster under the same column efficiency. Consumption of mobile phase declined 96% under same linear velocity.Using theμ-LC, chromatographic analytic methods of melamine, benzoic acid and sorbic acid in foodstuff were established. The retention time and the peak area RSD of each sample were less than 0.30% and 0.70%. Detection limits(S/N=3) were 0.015 mg/kg, 0.077 mg/kg, 0.084mg/kg respectively while linearly dependent coefficient were more than 0.999. Theμ-LC was also used to separate different samples including selfheal, PAHs and biogenic amines and the result was favorable.The construction and evaluation ofμ-LC has guiding significance to promote its industrialization and develop applied methods.
