Actinomycete SPRI-38490 which could produce herbicidal active metabolites was found by bioscreening. The strain was identified and its herbicidal metabolites were studied.The strain was identified by 16S rDNA gene sequence analysis and the study of morphological, physiological and biochemical characteristics. The results showed that the strain SPRI-38490 belonged to Streptomyces nodosus. With the help of silica gel chromatography, thin layer chromatography, and high performance liquid chromatography and other ways, a purified compound with herbicidal activities was isolated from the fermentation broth, named SPRI-38490-A. The spectra data of UV, MS,1H-NMR,13C-NMR and others were used to identify this compound. The molecular formula of the compound was confirmed as C23H29N5O11, with a molecular weight of 551. Compared with the known compounds in literatures and compounds database, the structure of SPRI-38490-A was as same as herbicidin A. Its herbicidal activities were proved. The reverse phase high-performance liquid chromatography analysis method was established, which could be used to analyse herbicicin A in the process of the fermentation.Compared with the known strains which could produce herbicidin A, the strain SPRI-38490 was different with any of them. The other strains produced other components of herbicidin while producing herbicidin A, but only herbicidin A was found in the metabolites of the strain SPRI-38490.
