Tarim red deer (Cervus elaphus yarkandensis) is a good subspecies of red deer population, it got it's name because it lives in the Tarim Basin from generation to generation. Tarim red deer can not only provide a bigger volume of production of pilose antler, but also provide ones with good quality.Tarim red deer which been fed in Xinjiang has a great number. As an antler strain with good quantity, the pilose antler of Tarim red deer is worthy of great efforts to develop. For a long time, when it come to talk about how to promote antler-industry of China, Chinese researchers have a common suggestion in establishing our own brand of pilose antler.To establish the brand of Tarim red deer, it's an important job for us to identify the pilose antler of Tarim red deer first, and to give a unique label to it. With the development of molecular biology techniques, as one of the molecular markers mtDNA has been widely used. 12SrRNA is part of the mtDNA sequences, which is a hot spot, is applied to the molecular phylogeny of animals, identification of animal source medicine and the study of deers. In this study, which the 12SrRNA sequences had been choosed, we found that the sequences differ significantly between Tarim red deer and the other 2 tested species. We designed a pair of allele-specific primers for diagnostic PCR and identified a distinctive site which can be digested by restriction endonuclease Psh B I on the 12SrRNA region. These tow techniques provide accurate, effective, and rapid approaches to Tarim red deer from the related subspecies.Pilose antlers only from sika deer(Cervus Nippon Temminck) and red deer(Cervus elaphus Linnaeus) are permitted by Provisions of the Chinese Pharmacopoeia. Not only in traditional medicine but also in modern pharmacological study pilose antler is thought to have strong efficacy in many ways.Because of good affection of pilose antler, investigators in many countries have a strong interest in study the secrets of pilose antler in modern research ways. They are analyzing the chemical composition of antler and looking for the unique pharmacological relationship between it's composition and efficacy, to contribute to the clinical applications of pilose antler. With the development of modern pharmacology the reason why antler can perform such a fast growth is that there is a natural growth factor database in pilose. Bone cells, nerve cells, fibroblasts and epithelial cell growth simultaneously in growth process of antler. Pilose antler, as the most active point of deer is a storage of a large number of growth factors: insulin-like growth factor (IGF), epidermal growth factor (EGF), nerve growth factor (NGF), transforming growth factor (TGF) and so on. They are biochemical basis of clinical effect, and have broad prospects in the clinical treatment.A decrease in bone-associated IGF-I levels could therefore result in remodeling imbalance and contribute to the gradual loss of bone that occurs with age. IGF-1 is used to prevent and reduce the loss of bone and treat osteoporosis. This is in accord with the strong efficacy of pilose antler in traditional medicine that antlers can strong bones and it may suggest IGF is the essential reason.EGF has promoting division and stimulation of anabolic effect on tissues developed from ectoderm and endoderm, so it can accelerate the healing of wounds and burns. This is in accord with the strong efficacy of pilose antler in traditional medicine and it may suggest IGF is the essential reason.Traditional processing of pilose antler will make it go through high temperature, which would cause damage to the active ingredients that can't stay at high temperature. Recent study suggest that it's better to select freeze-drying method to process pilose antler to make it become more effective in treating wound and strong bone.Therefore, we choose freeze-dried powder of antler as sample in this experiment, focusing on the IGF-1 activity and EGF activity in freeze-dried powder. In their cell proliferation experiments, aqueous extract of the freeze-dried powder significantly increased the proliferation of MCF-7 cell, and showing the IGF-1 activity; but the aqueous extract of freeze-dried powder didn't show a positive effect on Swiss 3T3 cell.
