The effects of ethanol concentration, solvent dose, ultrasonic time, ultrasonic power, and ultrasonic frequency on extraction yield were investigated. Diosgenin was extracted by ultrasonic extracting and hydrolyzing method from Dioscorea zingiberensis C.H.Wright. By the method of response surface methodology (RSM), the extration conditions were optimized. Two extracting solvents of water and 70 % (v/v) aqueous ethanol solution were used to obtain extractive substances (ES) from the Dioscorea Zingiberensis C.H.Wright. The kinetics of ultrasonic extraction was described by the unsteady-state diffusion through the plant material. Observed assemble behavior of the molecule of dioscin and diosgenin with atomic force microscope.The result showed that the optimum extraction conditions obtained for 10 g Dioscorea Zingiberensis C.H. Wright was as follows: ethanol concentration of 72 %, ethanol volume of 100 mL, ultrasonic time of 45 min and ultrasonic power of 420 W. The model obtained by RSM was well matched with practice conditions. Extraction yield of ultrasonic extraction method increased 26.46% compared with the traditional method. The coefficient of linear correlation was higher than 0.95. The maximum concentration of ES in the liquid extracts was reached after about 40 min of sonication. The data showed that the structure of dioscin and diosgenin is different: (1) At the density of 10-5 mol/L, the dioscin showed uniform small segment structure and analysis the molecule aggregate structure; But for the diosgenin, several molecules linked to form aggregated structure with the lower height and longer in length. (2) When the density reached to 10-4 mol/L, the dioscin appeared irregular sheet and small segment structure; the diosgenin was net structure in shape with many holes. (3) On the condition of 10-3 mol/L, the segment of dioscin assembled tightly on the mica; the diosgenin formed lager sheet structure with several small holes.The model was well matched with practice conditions. Extraction yield of ultrasonic extraction method increased 26.46% compared with the traditional method. The method is energy-saving, of low pollution, better efficiency and liability industrialization. The analysis of structure demonstrates that the molecules of dioscin and diosgenin formed different topography at different density. The molecule structure is the key factor for structures. The sugar chain of dioscin forced the molecules assemble not the small sheet structure. The hydroxyl group of diosgenin caused the molecules to link, forming the large sheet. The research of dioscin and diosgenin established the foundation for the interaction between drug and membrane.
