Celastrus angulatus Maxim. is a perennial woody vine plants of Celastraceae, Celastrus L. It has most development potential as one of the insecticides in China, also known as bitter bark, Luoshuang Red, Ma Duanchang and so on. Its roots, stems, leaves, fruits and seeds are not only important Chinese herbal medicine but also significant natural pesticides resources. As pesticides, its root bark, stem bark, leaves have been used to prevent various crop pests for a long time in civil. In view of the fact that celangulin possesses a series of different substitutedβ-dihydroagarofuan sesquiterpene polyolesters compounds of insecticidal active ingredient, and how to use them scientifically and effectively, this paper carried out a detailed study in the extraction process of the insecticidal active ingredient in the root bark of celangulin, in order to find an excellent extraction effect and feasible operation technology for insecticidal active ingredient. The main contents include:On the promise of knowing the structure and nature of angulatin A (celangulinV), which is the main insecticidal active component on toxicity in the root bark of celangulin, different origins of raw materials were analyzed firstly. Their contents were determined by HPLC. Considering that the source of raw materials was abundent, stability and convenience, Nanyang, Henan's was choosed. Then the six extraction solvents to celangulin were compared: methanol, ethanol, ethyl acetate, chloroform, benzene and petroleum. Comprehensive consideration of the extraction capacity, toxicity, economy and ease of recycling, methanol was choosed as the extraction solvent. And then the five extraction methods applying to celangulin were studied: hot reflux extraction of organic solvent (HFE), Soxhlet extraction(SE), microwave extraction(ME), ultrasonic extraction(UE) and supercritical CO2 fluid extraction(SFE). Based on viable economic principles in practical production process and the current trend of development, the experimental results showed that the simple and easy hot reflux extraction of organic solvent was the more ideal extraction method, which was suitable for laboratory and especially industrial. Based on the selection of raw materials source, extraction solvent and extraction method, in this part, it was experimented that the single-factor of the main factors: the extraction temperature, solid-liquid ratio, the extraction time and extraction number in extracting angulatin A, and obtained the optimum extraction conditions by orthogonal experiments: temperature, 50℃; feed ratio, 1:6; extraction time, 2 h; extraction numbers, 3. The results of the three parallel tests on the best conditions showed that the process was stable and feasible. The purpose of optimizing extraction conditions was attained. Taking account into the practical application of industrial production, the optimum conditions had been adjusted and amplified 100 times. During the scale-up experiments process, the results of its evaluation index: content of angulatin A, quantity of extract and composite index(quantity of extract x content of angulatin A), comformed with the small experiments. It could meet the need of industrial production requirement.
