| Gentiana macrophylla Pall, Gentiana straminea Maxim and Gentiana dahurica Fisch, belong to Gentianaceae Gentiana L Sect, as original gentian spices,were included in the "Pharmacopoeia of the People's Republic of China". The recent years'researches are focus on biological, chemical and pharmacological studies, but DNA fingerprints of the original plant of Qinjiao are rarely reported. On the basis of the the previous research, the subject, using both ISSR molecular markers and HPLC chromatographic methods, studied on Gentiana macrophylla Pall, Gentiana straminea Maxim and Gentiana dahurica Fisch,. The objectives were (1) to identify Gentiana macrophylla Pall, Gentiana straminea and Gentiana dahurica Fisch, and study the relationship between them. (2) to analyze the differences in gentiopicroside content between narrow-leaf and broad-leaf type of Gentiana macrophylla Pall for selecting the better gentiana herb resources.1.Inter-simple sequence repeat (ISSR) markers are employed to investigate DNA fingerprints of 13 populations of Gentiana macrophylla Pall, Gentiana straminea Maxim and Gentiana dahurica Fisch. A total 12 selected primers yielded 176 reliable bands including 168 polymorphic loci; there was one special band in G. macrophylla Pall, G. straminea Maxim and G. dahurica Fisch seperately. In the populations of Gentiana dahurica Fisch, only the population X-QQ had the band 358bp of the primer UBC810 to be different from others. There was abundant genetic diversity among the three species. The ISSR markers can be used for the identification of them each other.2. Both ISSR markers and HPLC methods were used to study on 13 populations of the three original plants (G. macrophylla, G. straminea and G. dahurica). Mantel tests showed the correlation of the clustering of the two methods were no significant (r= 0.160 63);The gentiopicroside content of them:G. macrophylla (an average of 80.481 mg/g)> G. straminea (an average of 68.600 mg/g)> G. dahurica (an average of 36.168 mg/g).3. The statistical tests showed that:the gentiopicroside content has a significant difference between the wide leaf-type and the narrow leaf-type G. macrophylla in underground portion. Because the later was significantly higher, it has the better quality; However,because of the significantly higher dry weight (biomass) of the former in underground part, then the output of gentiopicrin in the wide leaf-type was higher. So, the economic value of the wide leaf-type was higher too. The narrow leaf-type was significantly higher than the wide leaf-type in gentiopicrin content of aboveground parts. The difference was significant between the ground and underground parts in gentiopicrin content (t=30.492), then the gentiopicrin content of the later was far higher than the later. |
