Dryopteris crassirhizoma Nakai, a green perennial herb, bitter in taste, cold, slightly poisonous, was recorded in Shennong's Herbal the oldest book on Chinese traditional medicine and usually used for treatments of intestinal worms, bleeding and fever caused by influenza. Dryopteris crassirhizoma Nakai is a plant of genus Dryopteris in family Dryopteridaceae, and widely distributed in china, such as Heilongjiang, Jilin and Hebei provinces. But the wild resource is most abundant in Heilongjiang Province. It was reported that the rhizomes and roots in leafstalk of Dryopteris crassirhizoma Nakai have effects such as disinsectization, anti-tumor and anti-leukaemia. As a Chinese traditional medicinal herb, Dryopteris crassirhizoma Nakai has been paid more attention and it was used in clinic to cure anemopyretic cold, plaques heat rash, haematemesis, metrorrhagia and metrostaxis and other diseases.Because of complexity and polarity similarity of chemical compositions of the Dryopteris crassirhizoma Nakai, nothing but dryocrassin, filicic acids, aspidins, Dryopteris crassirhizoma glycosides, flavonoida, terpenoids and other enols were isolated. In order to further determine the chemical components of the plant, the antibacterial components and active sites of Dryopteris crassirhizoma Nakai were studied in this thesis. It was determined that n-butanol extracts were the active ingredients for Staphylococcus aureus and Bacterium coli resistant by turbidity method. Further studies of the active ingredients by Oxford cup method showed that 30% ethanol eluates were the active sites of antibacterial.30% alcohol eluates were further seperate by silicon column, ODS and HPLC and other updated isolation technologies. Thus 10 compounds were isolated by 1D-NMR and 2D-NMR technologies such as 1H-NMR,13C-NMR, HSQC and HMBC. And chemical structures of 5 compounds were identified. Study the antibacterial activity of 5 compounds obtained, the result shows that (-)-catechin has a strong antimicrobial effect.The studies on antibacterial activity compositions not only elaborate the effective antibacterial substance of Dryopteris crassirhizoma Nakai, but also provide valuable experiences for other dryopteris, and laid a foundation for development of high effect, low toxic, new medicine with determined ingredients and controllable and stable quality.
