Objectives:The incidence rate of gastric carcinoma is also in the first position among manlignant tumors in our country,and the mortality is about one quarter of global ones.Most patients have been progressing to advanced stage at the time of diagnosis. The rate of radical resection is lower, treatments including radiation, chemotherapy or preoperative chemotherapy have little infuence,even exert great side effect to patients.And at present having no normative chemotherapy, therefore the gene therapy has been introduced and displayed its promising. Two important properties of cancers are able to deregulate cell proliferation and suppress cell death, liver-intestine cadherin(CDH17) as one protein which have intimate correlation with invasion and metastasis of tumor,which as well as highly expresses in intestinalmucosa.This study extracted and identified CDH17/pcDNA3.1(-) eukaryotic expressing plasmid, and transfected it into BGC-826 cell by Lipofectmine 2000, the purpose is observing whether the extraneous CDH17 gene can transfect BGC-826 effectively as a anti-tumor factor. It provides the experimental foundation for further studying that transfection of this plasmid alone or combined with other plasmids to block the stomach neoplasms in different steps.Method:1 Transient transfection and expression of gastric tumor cellsGastric carcinoma BGC-826 cells were transfected with Lipofectamine 2000.Protein of cells were extracted at the time of 48h after tranfected, the method of Western blot was used to identify the expression of CDH17.2 The assays after transient transfection in vitroThe assays were divided into three groups,The cells transfected by CDH17/pcDNA3.1(-) was experimental groups,the cells transfected by pcDNA3.1(-) was blank control and BGC-826 cells untransfected was negative control. Cell matrigel invasion assay, scratching assay were used to study the effects of CDH17 gene.Results:1 Transient expression of CDH17. after transfected.After transfeted 48h, protein of CDH17 was detected with Western blot.2 All assays showed that the invasiveness and migration of BGC-826 cells transfected by CDH17/pcDNA3.1(-) were less than those of BGC-826 cells transfected by pcDNA3.1(-) and untransfected cells.Conclusion:CDH17 has been successfully transfected, they can be expressed in the gastric carcinoma cells.All assays suggest that CDH17 can affect biological characters of human gastric carcinoma cell line BGC-826. They may depress the invasion and metastasis of gastric carcinoma cell. Local cancer violations or distant metastasis often lead to treatment failure of the main reasons, the incidence of gastric cancer on the molecular mechanism of the biological behavior and the search for new anti-transfer measures is crucial.
