Object:To compare the impaired effectiveness of varous resuscitation fluid on morphology and function in hepatic cellular mitochondria of rats with hemorrhagic shock, to explore a more safe and effective resuscitation fluid for hepatic cellular mitochondrial impairment of hemorrhagic shock rats.Method:40 Wistar rats were randomly divied into 5 groups:Sham group(no hemorrhage, and no resuscitation),Shock group,lactated Ringers Solution(RL) group, hydroxyethyl starch (HES) group and autotransfusion(BL) group with eight rats per group. Each group was subjected to hemorrhagic shock for 1h.fellowed by resuscitation with lactated Ringers Solution, hydroxyethyl starch and blood for 2h. The rats were killed and their livers were sampled to measure values of dry to wet liver tissues weight ratio (D/W). After animal model was prepared, liver mitochondria were isolated as soon as possible.The activity of SDH, and the change of mitochondrial membrane potential were been tested, and ultrastructure of mitochondrial membrane was observed by electron microscope. The volume of bleeding and resuscitation fluid were recorded. All of the data were input into and analyzed by the database with the software of SPSS 16.0, The difference in groups was analyzed using one-factor analysis of variance. SNK—q test and chi-square test, P<0.05 for the statistical difference.Result:①The liver D/W ratio of the three resuscitation groups were significantly lower than that of the sham group(P<0.05), it in RL group were significantly lower than that of the HES and BL group(P<0.05).②In the three resuscitation groups, the ultrastructure of hepato cellular mitochondrial was damage obviously in RL group, and was impaired lower in BL group.③The activity of SDH in RL group and HES group were significantly lower than that of the sham group(P<0.05), but BL group was approached to sham group (P>0.05);The activity of SDH in BL group was significantly higher than that of the shock group(P<0.05); The activity of SDH in RL group and HES group were significantly lower than that in the BL group(P<0.05), but RL group was approached to HES group(P>0.05).④The change of mitochondrial membrane potential in RL group and HES group were significantly lower than that of the sham group(P< 0.05), but BL group was approached to sham group (P>0.05); The change of mitochondrial membrane potential in BL group was significantly higher than that of the shock group(P<0.05); The change of mitochondrial membrane potential in BL group and HES group were significantly higher than that in the RL group(P<0.05), but BL group was approached to HES group (P>0.05).⑤The bleeding volumes of resuscitation groups were approached to shock group (P>0.05). The resuscitation fluid volumes of BL group and HES group were significantly lower than that of RL group(P<0.05), but BL group was approached to HES group (P>0.05),Conclusion:This study confirmed that resuscitation can decrease impairment on morphology and function in hepatic cellular mitochondrial of rats with hemorrhagic shock, and the impaired degree in BL group was significantly lower than that in HES and RL group. With three different resuscitation fluid, HES group and BL group have the same effects, compared with lactated ringers solution, they can increase effective circulating blood volume and would be better fluid of resuscitation in rats with hemorrhagic shock. Accordingly, to transfuse autoblood is the most ideal resuscitation fluid for hemorrhagic shock.
