Objective: To study the chemical composition and total phenol quality control and the activity of scavenging free radical in vitro in H.rhamnoidesL. subsp. turkestanica Rousi in. Ann. native to Xinjiang. Methods: Use petroleum ether, chloroform, 70% ethanol, water to extract the chemical constituents of H.rhamnoidesL. subsp. turkestanica Rousi in. Ann. native to Xinjiang. Use silica gel, macroporous resin and sephadex LH-20 gel columniation chromatograp and crystallization methods to isolate compounds. Use chemical and spectral methods to identify the structures of the isolated compounds. Use F-C method and the orthogonal method screen the optimum extraction of total phenol. Use DPPH? method test the total phenol extraction to scavenge the free radical activeness in vitro. Use HPLC to determine the content of protocatechuic acid, rutin and isorhamnetin in the total phenol. Results: The seven compounds were elucidated as protocatechuic acid (Ⅰ), kinic acid (Ⅱ), rutin (Ⅲ), oleanolic acid (Ⅳ), Ursolic acid (Ⅴ),β-sitosterol (Ⅵ), daucosterol (Ⅶ). The total phenol optimum extraction and purification process : adding 30 times amont of 60% methanol, refluxing and extracting for 2 times, 3h each time the macroporous resin purification, 60% methanol elutions, the extract's total phenol content achieves 45.69%. The test of scavenge the free radical activity in vitro show the total phenol can scavenging the DPPH?, and the elimination ability higher than Vc. The HPLC method determined the protocatechuic acid, rutin and isorhamnetin in the total phenol extracts'total content is 13.5%. Conclusion: This research preliminarily determined the optimum extraction of the total phenol, establishes the method of total phenol content in H.rhamnoidesL. subsp. turkestanica Rousi in. Ann. native to Xinjiang for the first time. Test the total phenol, the protocatechuic acid, rutin and isorhamnetin scavenge the free radical ability in vitro. The HPLC preliminarily determined the protocatechuic acid, rutin and isorhamnetin in the total phenol extracts'total content.
