Cai Tony Tablets In Treatment Of Experimental Autoimmune Thyroiditis

Purpose:Taking experimental autoimmune thyroiditis of murine as model, thyroid peroxidaseantibodies(TPOAb)and thyroglobulinantibodies(TGAb) in blood,the structure of thyroid pathological tissue and the expression of TPO in thyroid pathological tissue by immunohistochemical staining method as index, to observe Pharmacological effects and mechanism of chaibei tablets to treat autoimmune thyroiditis.Material and method:An animal model of experimental autoimmune thyroiditis(EAT)in murine was developed by injecting thyroglobulin of porcine(PTg),mixed with Freund's adjuvant complete immunization in the first week to the third week and injecting thyroglobulin of porcine(PTg), mixed with Freund's adjuvant incomplete iimmunization in the forth week and the fifth week, in the meantime drinking Periodate water. We would divide them into bland comparing group,model comparing group,Chaibei tablets treating group and Tripterygium wilfordii Hook. f.. comparing group in the end of the fifth week. The blank comparing group and model comparing group would be given physiological saline, the treating group would be given Chaibei tablets, and the comparing treating group would be given Tripterygium wilfordii Hook. f..,giving medicine by irrigation stomach, twice everyday for four weeks continously. After dosing, we would take blood from orbitofrontal venous of the murine with capillary tube, and get blood serum by centrifugating to detect the level of thyroid peroxidaseantibodies (TPOAb) and thyroglobulinantibodies (TGAb) with chemiluminescence assay (CLIA). After getting the blood we would dissect to get the thyroids in the both sides after executing the murine by cervical dislocaitons method. The thyroids would be keeped by 10% Formaldehyde solution, to do pathology examination to the murine thyroid tissue, to observe the expression of TPO in thyroid pathological tissue by immunohistochemical staining method.Results:1. Serum findings said:Chaibei tablet treating group and Tripterygium wilfordii Hook. f. comparing group could both reduce the level of thyroid peroxidaseantibodies(TPOAb)and thyroglobulinantibodies (TGAb),Chaibei tablet treating group was compared with model comparing group P<0.05, tripterygium wilfordii Hook. f. comparing group was compared with model comparing group P >0.05, there was no statistics sense.2. Pathological findings said:Chaibei tablet treating group and Tripterygium wilfordii Hook. f. comparing group both had the pharmacological effect of protecting the structure of thyroid cells tissue and decreasing the infiltrating degree of inflammatory cells in thyroid tissue.3. Immunohistochemical findings said:Chaibei tablet treating group and Tripterygium wilfordii Hook. f. comparing group could both increase the number of TPO positive cells in thyroid tissue, and Chaibei tablet treating group was better than Tripterygium wilfordii Hook. f. comparing group.Conclusion:1. Chaibei tablet has the pharmacological effect of alleviating the level of thyroid peroxidaseantibodies (TPOAb) and thyroglobulinantibodies (TGAb) in serum of EAT murine.2. Chaibei tablet has the effect of protecting the structure of thyroid tissue of EAT murine.3. Chaibei tablet has the pharmacological effect of increasing the expression of TPO in thyroid tissue of EAT murine.
