| Purpose:To observe the effects of ChaiBei tablets on the experimentalautoimmune thyroid inflammation(EAT)rats from the aspects of thyroidantibodies,pathological morphology and the expression of cell apoptosisprotein(Fas,FasL),study the therapeutic effect of ChaiBei tablets and defineits pathogenesis from the aspects of apoptosis.Material and methods:90hea1thy female SD rats fed for oneweek(weight:160±210g)were randomly divided into6groups:normalgroup(G1):12;mode1group(G2):14;tripterygiumforrestii tablets treatmentgroup(G3):14;ChaiBei tablets Low dose-group(G4):12;ChaiBei tablets mediumdose-group(G5):12;ChaiBei tablets High dose-group(G6):14.Gl with commonfeed;G2-G6treatment:30mg Pig’s TG was completely dissolved in60ml PBSsolution,and15ml was thoroughly mixed with same volume complete freund’sadjuvant into water in oil type and gave subcutaneous injection0.2ml as primaryimmunization on back,abdominal,legs and neck of rats,need2times.Two weekslater,thoroughly mixed Pig’s TG15ml with incomplete freund’s adjuvant10mlas boosting immunization once a week combined with sodium iodide to feed therats with water until the seventh week(1L distilled water with0.64g sodiumiodide).From the eighth week,to feed G2with common feed and water;to feed G3with15mg/kg/d tripterygiumforrestii tablets by gastric perfusion;to feed G4-G6using respectively6g/kg/d,20g/kg/d,60g/kg/d ChaiBei tablets by gastricperfusion for four weeks.In thirteenth week,the rats were killed,blood and the thyroid tissues weretaken out with HE colored and observed the structure of rat’s thyroid tissueswith microscope. Thyroid apoptosis protein(TPOAb and TGAb)were tested by reagentbox’s show,Fas,FasL were tested by immunohistoehemistry.Results: 1.B1ood serum TGAb of model group increased and had significant difference(P<0.05) compared with the normal group; B1ood serum TPOAb of model groupincreased and had significant difference(P<0.01)compared with the normalgroup;Blood serum TGAb and TPOAb of tripterygiumforrestii group reduced and bothhad significant statistical difference(P<0.05)compared with model group;Bloodserum TGAb and TPOAb of formula ChaiBei tablets treatment groups reduced andhad statistical difference (P<0.05)compared with model group; Blood serum TPOAbof formula ChaiBei tablets treatment groups(hight group and low group)reducedand had statistical difference (P<0.05)compared with tripterygiumforrestiigroup;Blood serum TGAb of formula ChaiBei tablets treatment groups reduced andhad no statistical difference (P>0.05)compared with tripterygiumforrestiigroup.2. Pathological findings said Lymphocyte soakage degree of model groupIncreased and had significant statistical difference comparing with normalgroup;Lymphocyte soakage degree of tripterygiumforrestii group and ChaiBeitablets treatment groups decreased and had significant statistical differencecompared with model group. Lymphocyte soakage degree of and ChaiBei tabletstreatment groups decreased deeper thanrtripterygiumforrestii group.2.Immunohistochemical method findings said Fas,FasL immune pos1tive substanceof normal group presented light brown-red color,the expression ranges were verysmall;Fas,FasL immune positive of model group presented brown-red color,highconcentration and the expression ranges were extended compared with normal groupand OD value had significant differences(P<0.01);The color oftripterygiumforrestii group and ChaiBei tablets groups obviously faded and theexpression ranges of them became smaller and OD values had significantdifferences(P<0.05);Fas,FasL immune pos1tive substance and OD value of ChaiBeitablets groups had no significant statistical difference(P<0.05)compared withtripterygium-forrestii group. Conclusion:1. Chaibei tablets can significantly alleviate the levels of TGAb and TPOAb ofEAT rats.2. Chaibei tablets can reduce lymphocyte inflammatory cells soakagedegree,lighten its,inflammatory response.3. ChaiBei tablets can effectively restrain the apoptosis thyroid epithelialcells and reduce over activation. |
